| Background: With the rapid development of China’s economy,enterprises and factories have been increasing,and some safety accidents have also occurred frequently,which has increased the incidence of spinal cord injuries.However,rehabilitation after spinal cord injury is a major medical problem in the world today.There are currently no effective drugs and surgical treatment methods.Therefore,it is of great scientific value and practical significance to explore the methods of spinal cord rehabilitation.Previous studies have shown that it is possible to promote the repair and regeneration of spinal cord neurons through the use of electrical acupuncture.On the other hand,rehabilitation training at the late stage of surgery can also greatly help rehabilitation after spinal cord injury.However,these two treatment methods are still controversial,and there are few studies on electroacupuncture combined with sports rehabilitation training.Objective: This study is to confirm the beneficial effects of Du meridian electroacupuncture and treadmill training on functional recovery of spinal cord injured rats,and to study the effects of Du meridian electroacupuncture combined with treadmill training on functional recovery of spinal cord injured rats and the related regulatory mechanisms.Methods: Purchase 65 healthy adult rats from Kunming Medical University Laboratory Animal Center,weighing 260g-280 g.First,the rats were screened and 50 healthy individuals were screened to walk successfully on the running platform.Fifty rats were randomly divided into groups;Sham,Control,Electroacupuncture,Treadmill,Electro-acupuncture and Treadmill.The control group,electroacupuncture group,treadmill training group,and electroacupuncture combined treadmill training group were used to make spinal cord injury models.The experimental time is five days a week for seven weeks.Weigh the body once a week,measure the urine volume,do a BBB score,a grab test,a tail-flick test,and a slanting plate test.Then MRIdetection,pet-ct detection,electrophysiology,material extraction,and frozen storage were performed.Finally,the data was analyzed with SPSS21.0 statistical software.Results: The experimental results show that(1)body weight and urine output:Compared with the control group,the body weight of the electroacupuncture group,treadmill training group,and electroacupuncture combined treadmill training group increased significantly,and the difference was statistically significant(P <0.05).Compared with the electroacupuncture group and the treadmill training group,the weight of the electroacupuncture combined treadmill training group increased significantly,and the difference was statistically significant(P <0.05).(2)Temperature and pain: Compared with the control group,the temperature and pain of electroacupuncture group,treadmill training group and electroacupuncture combined treadmill training group were significantly improved,and the difference was statistically significant(P <0.05).Compared with the electroacupuncture group and the treadmill training group,the acupuncture combined with the treadmill training group significantly improved the pain and warmth,and the difference was statistically significant(P <0.05).(3)Behavior: The recovery of hind limbs in the control group was not obvious,the recovery in the treadmill group and the electroacupuncture group was better,and the recovery in the electroacupuncture combined treadmill group was the best.(4)In terms of imaging: DTI results showed that compared with the Sham group,the spinal nerve fibers in the control group were significantly reduced,and the difference was statistically significant(P <0.05).The electroacupuncture group,treadmill group,and electroacupuncture combined treadmill the number of nerve fibers in the group increased significantly compared with the control group,and the difference was statistically significant(P <0.05).PET / CT results showed that compared with the Sham group,the absorption values of spinal cord and lower limb gastrocnemius muscles in the control group were significantly reduced,and the difference was statistically significant(P <0.05).Compared with the control group,the electroacupuncture group,treadmill group and the absorption value of the acupuncture combined treadmill group increased significantly,and the difference was statistically significant(P <0.05).(5)Electrophysiology: The results of exercise-evoked potential showed that the amplitude and area of the electroacupuncture combined treadmillgroup were significantly higher than those of the control group,and the difference was statistically significant(P <0.05).The electromyographic results showed that the amplitude and area of the electroacupuncture combined treadmill group were significantly higher than those of the control group,and the difference was statistically significant(P <0.05).(6)Morphology: Immunofluorescence showed that the number of microglia in scars of spinal cord injury rats in electroacupuncture group and treadmill group was significantly reduced compared with the control group,and the difference was statistically significant(P<0.05).Compared with the electroacupuncture group and the treadmill group,the number of microglial cells in the scar of the electroacupuncture combined with treadmill group rats was significantly reduced,and the difference was statistically significant(P <0.05).Conclusions: Electro-acupuncture improved functional recovery in rats with spinal cord injury.Treadmill training is also effective in functional recovery of spinal cord injured rats,and it is mostly reflected in behavioral aspects.Electro-acupuncture combined with treadmill therapy has a very good therapeutic effect on the functional recovery of spinal cord injury rats.Compared with the control group,it has significance in the entire treatment process.Compared with the single treadmill and electro-acupuncture alone,there are statistics in the first three weeks.Significant but lost statistical significance after four weeks but its recovery trend was significantly better than that of the treatment alone group. |
