Chronic atrophic gastritis(CAG)is one of the most common digestive system diseases worldwide which defined by WHO as initial step of cancer.Gastrodia elata Blume(GEB)is a traditional herbal with multiple pharmacological activities which was widely used in Asian countries.The aim of this study was to investigate the effects of autoimmune-induced chronic atrophic gastritis on metabolites in rats and to evaluate the preventive and therapeutic effects of Gastrodia elata on chronic atrophic gastritis.In this study,rats were divided into six groups: blank control group(CON,saline),model group(MCG,saline),prophylactic administration group(PGE,0.4g/kg),high dose group(HE,0.4g/kg),low dose group(LGE,0.1g/kg)and positive drug group(DOM,1mg/kg).For the first time,we used 1H NMR-based metabonomics with histopathological examination to analyze the stomachs of rats.The biochemical indexes of MDA,SOD,GSH,NO and XOD were measured.The results showed that GEB had a significant therapeutic effect on chronic atrophic gastritis at the dose of 0.4g/kg.Gastrodia elata Blume could apparently ameliorate the damaged gastric glands and the biochemical parameters,enhance gastric acid secretion,and significantly relieve the inflammation of the stomach.Orthogonal signal correction-partial least squaresdiscriminant analysis(OSC-PLS-DA)of NMR profiles and correlation network analysis revealed that Gastrodia elata Blume could effectively treat CAG via regulating energy and purine metabolisms,and by anti-oxidation and anti-inflammation effects.In the experiment of evaluating the pharmacodynamics of different polar parts,Gastrodia elata was divided into four polar parts: water phase,n-butanol phase,ethyl acetate phase and petroleum ether phase.The therapeutic effect of different polar parts on chronic atrophic gastritis was evaluated at a dose of 0.4 g/kg.Metabonomic analysis showed that the extracts from water phase and n-butanol phase of Gastrodia elata had better pharmacodynamic effect.Therefore,we speculated that the antioxidant and anti-inflammatory components of Gastrodia elata were concentrated in the large polar part.
