| Objective: To observe the relationship between SLCO1B1 gene polymorphisms and serum lipids in patients with type 2 diabetes mellitus(T2DM)in Zunyi area,and to explore the relationship between SLCO1B1 gene polymorphisms and serum triglycerides(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C),which provide new precision medicine and safe medication for Han T2 DM population in Zunyi area diagnosis and perspective treatment.Methods: Randomly select 170 Han T2 DM patients from July 2018 to January 2019 who were admitted to the General Outpatient Clinic and Physical Examination Center of the Affiliated Hospital of Zunyi Medical University,confirmed by medical history and laboratory examinations,and after followed-up all their parents were Han.According to the 2016 "Guidelines for the Prevention and Treatment of Dyslipidemia in Chinese Adults",the selection criteria for the hyperlipidemia group was defined as TC≥5.6 mmol/L,which was divided into the normal lipid group(TC<5.6 mmol/L,N = 90 cases)and hyperlipidemia Syndrome group(TC≥5.6mmol/L,N=80 cases).All enrolled subjects were measured for age,sex,height,weight,and calculate body mass index(BMI),determine SLCO1B1 genotype,fasting plasma glucose(FPG),TG,TC,HDL-C,LDL-C by laboratory inspection.Compare FPG,TG,TC,HDL-C,LDL-C,SLCO1B1 388A>G and 521T>C genotype differences in normal blood lipid group(TC<5.6mmol/L)and hyperlipidemia group(TC≥5.6mmol/L).Factors with statistically significant differences in univariate analysis were incorporated into a multivariate logistic regression model to evaluate the independent association of genotype groups with serum lipid levels in T2 DM patients.Factors with statistically significant differences in univariate analysis were incorporated into a multiple linear regression model to evaluate the independent association of genotype groups with serum lipid levels in T2 DM patients.Results: Compared with the normal lipid group,the levels of FPG,TC,TG,LDL-C,HDLC in the dyslipidemia group were significantly increased(P<0.05);there was no significant difference in age and BMI between the two groups(P>0.05).At the 388A>G locus of the SLCO1B1 gene,there was no statistically significant difference in the distribution of genotypes between the hyperlipidemia group and the normal blood lipid group(P>0.05).At the 521T>C locus of SLCO1B1 gene,there was no statistically significant difference between the CC group and the TC group in the hyperlipidemia group and normal blood lipid group of T2 DM patients;the CC group and the TT group,the TC group and the TT group the distribution of the type group in T2 DM patients with hyperlipidemia group and normal blood lipid group was statistically significant(Bonferron correction,P<0.0167).The SLCO1B1 genotype of the T2 DM population was divided into two loci of 388A> G and 521T>C.The genotypes of the two loci were determined and the Hardy-Weinberg genetic balance test was performed.There was no significant difference in the number of statistical cases(P>0.05),consistent with Hardy-Weinberg’s law of genetic balance,indicating that the 170 selected T2 DM patients had no mating relationship and no genetic drift,and were representative in Zunyi area.The serum TC,TG,HDL-C,and LDL-C levels were compared at the 388A> G and 521T> C loci of the SLCO1B1 genotype,and the TG and TC between the genotypes at the 388A>G locus were obtained.,HDL-C and LDL-C levels were not significantly different(P>0.05).At 521T>C,the TC and LDL-C levels of CC genotypes were higher than those of TT genotypes and TC genotypes,and the differences were statistically significant(P<0.05);the TG and HDL-C levels were different among the genotypes.No statistical significance(P>0.05).Conclusion: The 521T>C locus CC allele of the SLCO1B1 genotype of Han T2 DM patients in Zunyi area is associated with abnormally elevated levels of serum lipids,TC and LDL-C,but it is not an independent risk of abnormally elevated TC and LDL-C levels factor.And 388A>G polymorphisms have nothing to do with blood lipid levels.The determination of SLCO1B1 genotype control of dyslipidemia and the reduction of the risk of complications in the treatment of T2 DM patients may have a certain effect. |
