| Objective:To establish a comprehensive evaluation method for the quality of the Rheum Radix et Rhizoma fresh-cut pieces.To optimize the best process of the Rheum Radix et Rhizoma fresh-cut pieces.To compare and analyze comprehensively and systematical-ly fresh-cut pieces and traditional pieces,and compare the quality difference between The two pieces,providing a basis for the feasibility study of Rheum Radix et Rhizoma fresh-cut.To study the quality standard of Rheum Radix et Rhizoma fresh-cut pieces,and draft preliminarily a draft quality standard,which provides a theoretical basis for the quality control for them.Methods:Using HPLC to simultaneousy determine the content of 11 ingredients in Rheum Radix et Rhizoma,combing with water-soluble extracts,decoction appearance characteris-tics and forming rate,to establish a comprehensive evaluation method for the quality of the Rheum Radix et Rhizoma fresh-cut pieces and comprehensively evaluate its quality.Taking the comprehensive score as the inspection index,the fresh cut degree,the cutting specifications and the drying temperature are the investigating factors,and the Box-Beh nken response surface testis used to design and optimize the process parameters of Rheum Radix et Rhizoma.Adopt the comprehensive technology and method of“appear ance traits component analysis pharmacological and medicinal effects"from the aspects of chemical ingredients,water-soluble extracts,appearance traits and diarrhea,anti-inflamat ory,etc,comprehensively and systematically analyze the fresh-cut processing thchnology and traditional processing,compare the quality difference between the two pieces.Accor-ding to the detection method of the 2015 edition of"Chinese Pharmacopoeia",the cha-racteristics,identification,inspection and determination of extracts of Rheum Radix et Rhizoma decoction pieces are detected.the fingerprint is established.Results:1 The chromatographic condition for the simultaneous determination of 11 compo-nents in Rheum Radix et Rhizoma fresh-cut pieces was established by HPLC:Agilent HC-C18?2?column?4.6 mm×250 mm,5?m?,0.1%phosphoric acid methanol as mobile phase,gradient elution,015 min,40%50%methanol,1530 min,50%60%methan-ol,3050 min,60%80%methanol,5080 min,80%100%methanol,volume flow rate1.0 mL·min-1,detection wave length 410 nm,column temperature 30?,injection volume10?L.2 Established a method to comprehensively control the quality of Rheum Radix et Rhizoma fresh-cut pieces with free anthraquinone derivatives,combined anthraquinone derivatives,water-soluble extracts,appearance characteristics and forming rate of pieces.The index weight coefficients are set to 0.2,0.1,0.2,0.25,and 0.25 respectively,and the degree of membership=?index value-minimum value of index?/?maximum value of inde-x minimum value of index?,comprehensive score=product of the degree of membership of each index and weight coefficient Sum.3 Established the best fresh-cut processing thchnology of Rheum Radix et Rhizo-ma:Fresh Rheum Radix et Rhizoma removes soil,fibrous roots,and skins.According to the different shapes,cut the petals or segments,process into Suji or Dan Ji,set in a bla-st drying oven,dry at 40?to 35%moisture content,cut into 2.5 mm thick pieces, and put into the blast drying oven Dry at 60?.4 There are some differences in the appearance characteristics,forming rate,water soluble extract and chemical composition of the three batches of Rheum Radix et Rhi-zoma fresh-cut pieces and traditional pieces.The overall performance is that Rheum Radix et Rhizoma and traditional pieces in diarrhea and anti-inflammatory.Diarrea effect results show the fresh-cut pieces group had a clearer effect than the traditional pieces group,comparing with the traditional pieces group.The anti-inflammatory results show differe-nt pieces have anti-inflammatory effects.The inhibition rates of xylene induced mouse ear swelling in high and low-dose groups of fresh-cut pieces,traditional pieces high and low-dose groups,and aspirin groups were:74.30%,39.54%,67.05%,33.94%,82.81%.6 Fresh-cut pieces of Rheum Radix et Rhizoma are irregular and thick round sli-ces,about2.5 mm in size.TLC identification,at the position corresponding to the control medicinal material chromatography,the same five orange-yellow fluorescent main spots were shown.at the quality of Rheum Rad-ix et Rhizoma fresh-cut pieces can comprehensively evaluate their quality,and provide a basis for their the optimization of processing technology.2 The processing technology of the optimized fresh-cut pieces is stable,simple and reliable,scientific and reasonable,which ensures the stable and controllable quality of The pieces,and can provide new ideas for the production of
