Objective:(1)Study the distribution characteristics of LHCGR gene rs7579411 locus polymorphism in Guangxi population and compare the distribution differences among different ethnic groups.Compare the differences between LHCGR gene rs7579411polymorphism in patients with PCOS and controls.(2)To investigate the relationship between rs7579411gene polymorphism and PCOS endocrine and metabolic indicators and subtype in Guangxi population.Methods:Selected 573 healthy patients as normal group in part one,the im LDRTMSNP was used to detected the genotypes of LHCGR gene rs7579411,and the DNA sequencing was used to validated the genotyping results.To compare the polymorphism data of LHCGR gene among Beijing population(Hap Map-CHB),Japanese population(Hap Map-JPT),European population(Hap Map-CEU)and African population(Hap Map-YRI)published by the Hap Map.A total of 234 PCOS patients and 248 normal patients of similar age were selected as control subjects in the second part,the im LDRTMSNP was used to detected the genotypes of LHCGR gene rs7579411,calculate the genotype and allele frequency of the two groups,compare the distribution of allele and genotype frequency in the PCOS patients and contrast groups,comparison of gene polymorphisms between PCOS and control groups in the BMI、LH/FSH、T、AMH of endocrine metabolic indexes.To calculate the genotype and allele frequencies of A、B、C and D four subtypes,comparison of genotype and allele frequencies of rs7579411 loci between four subtypes and control groups.Results:(1)The rs7579411 loci in Guangxi population contained CC、CT、TT genotypes,with C/T genotypes more common(47.1%)and alleles more common with C(62.7%).There was no significant difference in the distribution of genotype and allele frequency between male and female rs7579411(P>0.05).(2)The genotype frequency of population rs7579411 in Guangxi was statistically significant compared with Hap Map-JPT、Hap Map-YRI(P<0.05),but not statistically significant compared with Hap Map-CHB、Hap Map-CEU(P>0.05).The allele frequency of rs7579411 sites was statistically significant compared with Hap Map-JPT、Hap Map-YRI(P<0.05),but not statistically significant compared with Hap Map-CHB、Hap Map-CEU(P>0.05).(3)There was no significant difference between genotype and allele frequency of rs7579411 locus in PCOS and control group(p>0.05).(4)rs7579411 locus CC、CT and TT genotypes were no statistically significant compared with BMI、LH/FSH、T、AMH indicators(p>0.05).(5)There was no significant difference between genotype and allele frequency of LHCGR gene rs7579411 in A、B、C and D four subtypes and control groups(p>0.05).Conclusion:(1)The distribution frequency of genotype and allele of LHCGR gene rs7579411 locus was not related to sex in Guangxi population,(2)LHCGR gene rs7579411locus polymorphisms differ from other races.(3)There was no significant difference between genotype and allele frequency of rs7579411 loci in PCOS and control groups(p>0.05).(4)There was no significant correlation between rs7579411 locus polymorphism and PCOS pathogenesis.(5)LHCGR gene rs7579411 SNPs was not significantly associated with the four subtypes and the control group.
