Objective:Cinnamomi cortex,a common warm medicine of the traditional Chinese medicine,which can be used for functional food,and whose clinical efficacy was affected by its quality directly.A unified quality evaluation system to evaluate its quality was urgent to developed to guide its classification.Therefore,it is necessary to establish a comprehensive quality evaluation system to provide a reference for high quality and high price.Method:In this paper,based on the extensive collection of cinnamomi cortex from the main production areas in China,according to Chinese Pharmacopoeia(2015 edition),45 batches of cinnamomi cortex have been checked on the related regulations of cinnamomi cortex item.Referring to the thin layer chromatography conditions of cinnamomi ramulus in United States Pharmacopeia,and the analysis of chemical components of cinnamomi cortex on previous research,an optimized TLC method was developed for the identification of cinnamaldehyde,coumarin,cinnamyl alcohol,cinnamic acid and2-methoxy cinnamaldehyde in cinnamomi cortex,which can effectively distinguish the cinnamomi cortex from its adulterants,cinnamomum zeylanicum and cinnamomum burmann.Meanwhile,a reliable and feasible strategy of improvement on quality standard of cinnamomi cortex were developed and validated by modern technology,including high-performance liquid chromatography(HPLC),Fingerprint and Quantitative Analysis of Multi-components by Single-marker(QAMS),which promote the improvement quality control level of cinnamomi cortex.The chemical quality control system of "the combination of multi-component content determination and fingerprint" were adopt,and11 index components such as Cinnamaldehyde,were selected to establish the method for the quality control of cinnamomi cortex.A reliable and feasible strategy of chromatographic fingerprinting was developed and validated by ultra-high performance liquid chromatography coupled with diode array detection,which would be a useful reference for the development and further study of cinnamomi cortex.Furthermore,to identify more meaningful chemical markers,Chemometrics methods including similarity evaluation for chromatographic fingerprinting of TCM,principal component analysis(PCA),hierarchical clustering analysis(HCA)were further to applied to analyze the data.Result:1.According to the 2015 edition of the Chinese Pharmacopoeia,the source,characters,microscopic and TLC identification,moisture and total ash examination,determination of cinnamaldehyde and volatile oil were carried out on 45 batches of cinnamomi cortex.The results showed that all the 45 batches of cinnamomi cortex,except one batch of cinnamomi cortex with cinnamaldehyde unqualified,met the requirements of the Chinese Pharmacopoeia(2015 edition),which provided sufficient sample information and representativeness for the establishment of the quality evaluation standard of cinnamomi cortex.2.The contents of 11 chemical constituents,such as cinnamyl alcohol,cinnamic acid and cinnamaldehyde,were determined by UPLC.The results showed that the contents of 11 active components in cinnamomi cortex from different habitats were different,and cinnamaldehyde was the highest constituents.At the same time,considering the cost of analysis and detection,a quality control method of quantitative analysis of multi-components by single-marker for four active components in cinnamomi cortex was established by using cinnamic acid as internal reference material,which has high content and low price.The results showed that the quantitative analysis of multi-components of cinnamomi cortex by single-marker was consistent with the multi-component analysis by external standard,indicating the quantitative analysis of multi-components by single-marker was effective and feasible for the quality evaluation of cinnamomi cortex.3.In this paper,UPLC technology was used to construct the fingerprint of cinnamomi cortex for comperhensive evaluation,9 characteristic peaks were chosen,and 5 of them were identified.It was uesd to distinguish cinnamomi cortex from from its adulterants,cinnamomum zeylanicum and cinnamomum burmann.Furthermore,the chemometrics methods including hierarchical cluster analysis(HCA),principal component analysis(PCA)and similarity,which all generate quality evaluations and correlation classifications of cinnamomi cortex,were introduced to improve the cinnamomi cortex quality control standards.It is well divided into Guangdong and Guangxi,and geographical differences were very revealing in the results of classification.4.The fingerprints of cinnamomi cortex with different growth years were studied by UPLC.A comprehensive evaluation of cinnamomi cortex with different growth yearwas developed by the chemometrics methods including hierarchical cluster analysis(HCA),principal component analysis(PCA)and similarity.The results showed that there were significant differences in the active components of cinnamomi cortex between over 30 years and under 30 years,especially in the contents of coumarin,2-hydroxy cinnamaldehyde and cinnamic acid.The content of volatile oil in cinnamomi cortex was determined by the method of water distillation,indicating the content of volatile oil in cinnamomi cortex increased gradually with the increase of growing years.Conclusion:The modern scientific and technological means(Ultra performance liquid chromatography technology,multi-evaluation and fingerprint technology),combined with a variety of modern statistical methods(including similarity evaluation,cluster analysis and principal component analysis)were developed to establish the feasible scheme of improving the quality standard of cinnamomi cortex,which not only can improve the technical level of quality control of cinnamomi cortex,but also can provide sufficient data support for establishing the specification grade standard of cinnamomi cortex.It provides a systematic and applied research foundation for setting the price according to quality.
