Studies On Chemical Consitituents And Improving Quality Standard Of Curculiginis Rhizoma

Curculiginis Rhizoma is the dried rhizome of Curculigo orchioides Gaertn(Hypoxidaceae family),it has been used for impotence,limb limpness and arthritis of the lumbar and knee joints.“Xianmao” has a long history of medicinal use and has been documented in successive literatures.In this paper,based on the chemical constituents of Curculiginis Rhizoma,combined with the pharmacological effects and clinical studies,a newly component orcinol glucoside were added as the reference substance.the quality standard of Curculiginis Rhizoma was improved comprehensively,and the content of orcinol glucoside and curculigoside in Curculiginis Rhizoma was determined.Fifteen compounds were obtained by using modern separation technologies.Their structures were identified by spectroscopic analysis,polarimeter and single crystal diffractometer.They were 3 lignans,3 terpenoids,9 phenolic acids and phenolic glycosides,of which 4 new compounds including Curculigine P,Curculigine Q,1-Oethoxylcurculigine and curculigoside J were isolated and 4 compounds including nyasicol,3??-dehydroxynyasicoside,nyasicoside,and limonin were isolated from the species of Curculigo orchioides for the first time.The isolated compounds were determined on 5?-reductase activity,as a result,curculigoside,orcinol glucoside,orcinol gentiobiose showed 5?-reductase promoting effect and 2,6-methoxyphenol,nyasicol,and 3-dehydroxynyasicoside showed 5?-reductase inhibiting effect.According to our reseach results and present clinical research reports,orcinol glucoside was suggested to add as a qualitative and quantitative indicator of Curculiginis Rhizoma.The TLC identification method of Curculiginis Rhizoma in the Chinese Pharmacopoeia(2015 edition)was revised with a mixture solution of dichloromethane,acetone and formic acid(5:2:1)as mobile phase and a solution of 2% vanillin sulfuric acid as the derivatized reagent.A method for determination of orcnol glucoside and curculigoside by High pressure liquid chromatography(HPLC)was established with the same test solution preparation and different chromatographpic conditions.Totally,the contents of twenty-one batches of Curculiginis Rhizoma samples from different places were determined.The average contents of orcinol glucoside and curculigoside were separately 7.636 and 0.541 mg/g.According to the content requirement of curculigoside in Chinese Pharmacopoeia of 2015 edition,only two batches were qualified.It is speculated that the reason may be due to the difference of planting environment,harvesting time and growing years of the plant.
