Based on literature data, related specimens, commercial samples and field investigation, botanical origin of "xianmao" was clarified. They was found that the rhizome of "xianmao" used in the traditional Chinese drugs in China from Tang dynasty on (recorded in Hai YAO Ben Cao) was derived from Curculigo orchioides Gaertn. of Hypoxidaceae, only species of Curculigo Gaertn. used in China.The distribution of genus Curculigo Gaertn. ,characteristics of plant, tissues and cells character, power character of rhizome, the related stereological parameters of tissues and cells By computer image analysis, RAPD markers were researched for the first time. Then combined with numerical taxonomy and principal component analysis, its features and standards for the comprehensive evaluation were illustrated qualitatively and quantitatively. The varieties of Curculigo Gaertn. were generally distinguished at the some threshold level of clustering distance. The classification of the genus Curculigo Gaertn. in China by The Integrated evaluation qualitatively and quantitatively were illustrated. These new approach of species recognition is scientific, and practical, and is especially suitable for the identification of those that are easy-confused or of close affinity.The Percentage of similarity to the Bray—Curtis distance coefficient of community plot was carried out to divide the community types of Curculigoorchioides Gaertn. The community of Curculigo orchioides Gaertn. can be divided into five types: Pinus massoniana Lamb, community, Castanea mollissima Bl. community, Cyclobalanopsis spp. or Quercus spp. community, Pinus massoniana Lamb.+Cunninghamia lanceolata (Lamb.)Hook.—Dicranopteris dichotoma (Thunb.) Bernh. community and grasslands by the percentage of coverage. The distribution pattern of Curculigo orchioides Gaertn. population was studied by varied scale of observation—according to testing frenquency distribution and patchiness indices such as C,K,m*/m, I. the distribution pattern of Curculigo orchioides Gaertn. was mainly aggregation distribution.The influencing factors of ISSR and RAPD were analyzed by adjusting the concentration of template DN A, Mg2+ , dNTPs and primer, the contents of Taq DNA polymerase etc. The stable and reproducible optimum reaction system of ISSRand RAPD------PCR amplification and PCR amplification parameters wereestablished for Curculigo orchioides Gaertn.To take up the systematic diagram of genetic relationship by calculating the proximity coeficient and cluster analysis. The selected random primers used in PCR amplification to produce polymorphic bands in Curculigo orchioides Gaertn. samples. Results of cluster analysis show that its population can be divided into three group according to the genetic diversities and genetic relation between different population, which proves that it is feasible to differentiate the genetic population.
