Curculigo is a perennial plant, which is the dry rhizome of Curculigo orchioides Gaertn.It is included in the Chinese pharmacopeia. As a traditional Chinese medicine,it have been mainly used for treatment of impotence,enuresis, cold sperm, cold pain of back and knee, numbness of the limbs, and decline in physical strength. Curculigo widely distributed in Asia, Afirca, South America and the oceaniantriopical and subtriopical areas.Our country have7, is mainly produced in southwest and southern China. For all I know, Curculigo orchioides Gaertn has been in wide state. And in recent years, with the improvement of people’s living standard,the demand for Curculigo is more and more,but it was not to belong to arundinacea the national protected plants, so its protected degree is very limited.From the60s, domestic and foreign scholars had began carried on the research of the chemical composition of Curculigo orchioides Gaertn.So far, the main constituents of Curculigo orchioides Gaertn. are phenol and phenlic glycoside, chlorine phenol, triterpene saponin, lignin and lignin glycoside, flavones, etc. Primise research shows that:90%ethanol extracts of Curculigo orchioides have cell toxicity and resistance to MRSA activity.To reveal the material base of its cell toxicity and anti-MRSA activity, We had a separation and identification in the90%ethanol extracts of Curculigo orchioides with the different conventional chromatography and all compoundswere identified.Then we have a determination in the their cell toxictity and anti-MRSA activity. We had obtained seven compounds from Curculigo orchioides Gaertn.and identified six known compounds through the determination of various spectrum ananlytical and synthesis and the compounds physical, chemical properties,chemical constant and They are (3-sitosterol(1),2,6-Dimethoxybenzoic acid(2), Daucosterol(3), Curculigoside(4),Orcinol glucoside(5) and orcinol(6).For the first time, compound4,5,6were studied in the inhibition of HepG2cell growth(MTT method) and anti-MRSA (trace broth dilution method) activity.The result is that compound4,5and6inhibit the growth of HepG2IC50were134.9,221.4and144.4μg.mL-1.MIC of MRSA of the compound4,5and6were>1024,>1024、128~256μg.mL-1. The results show that the compound4,5and6have a certain cell toxicity in the HepG2cells, and the compound6have a certain anti-MRSA activity. In addition, in the view of compounds of4,5and6have a certain cell toxicity and anti-MRSA activity, and the structure of compound2is approximate with the compounds of4,5and6.They are the main constituents of Curculigo orchioides, For the first time, Our subject is used the compounds of2,4-6as the index component and carried out the fingerprint research about10samples were collected from the different region in China.At the same time, it has been established to simultaneously the quantify of the four consitituets in Curculigo orchioides (such as Orcinol glucoside,orcinol,2,6-Dimethoxybenzoic acid, Curculigoside,) It’s a rapid, sensitive and accurate HPLC-DAD analytical method and can be used as the reference of the quality evaluation methods of Curculigo orchioides Gaertn. The results revealed that the chemical constituents of these samples were obviousely different,and the four consitituets content also differed in these samples.
