Objective Detection of the elderly cognitive impairment in patients with estrogen receptor alpha?ER-a?gene promoter methylation status and the same type of homocysteine?Hcy?,estradiol?E2?level and compared with normal people,to investigate the promoter methylation status and Hcy geriatric cognitive disorders and ER alpha gene promoter,ER alpha correlation.Method According to the formula of epidemic survey,Baotou City,the sixth census data and the cognitive impairment prevalence,estimated in Baotou,Inner Mongolia region required survey sample of 384 Cases,using multiple stratified cluster random sampling method,using the MMSE,MOCA equivalent scale screening,confirmed 54 cases of elderly cognitive disorder patients as case group,and randomly selected 108 cases of healthy people as control group.Venous blood of all the research object,using fluorescent biochemical method and chemical chemiluminescence detection of Hcy and serum levels of estradiol?E2?,using the NMSP method was used to detect the ER alpha gene promoter methylation of,by real-time The PCR method was used to detect the expression of ER-alpha m RNA,and all the data was analyzed by SPSS 17.Results?1?There were no significant differences in age,sex composition and BMI between two groups.The cognitive impairment group was compared with the control group.?2?the level of E2 in the case group was lower than that of the control group?P<0.05?,the differences were statistically significant.?3?Case group of 54 specimens with 37 cases of methylation,the methylation rate was 68.5%.in the control group?n=108?in 28 cases of methylation,the methylation rate was 25.9%.By chi square test,the case group and the control group methyl the rate difference was significant?P<0.05?.In the second group sex between a comparison group of rate had no difference?P>0.05?.?4?Cases of ER-?m RNA]CT value 14.31+1.63,control group]CT value was 10.50+1.22,between the two groups in the m RNA expression of the deletion rate was 0.052?P<0.01?,the difference is statistically significant.?5?After the Spearman rank correlation analysis,case group ER alpha gene start promoter methylation and m RNA expression of deletion??CT?correlation coefficient is r=0.878?P<0.05?.And with the increase of?CT value and degree of methylation also delivery time increased.case group ER alpha gene promoter promoter region methylation degree and t Hcy correlation coefficient r=0.552?P<0.05?.And with elevated t Hcy concentrations and degree of methylation also delivery time increased.case group ER alpha gene start promoter region methylation degree and E2 had no obvious correlation,r=0.077?P>0.05?.Conclusions?1?The ER-?gene promoter Cp G Island promoter hypermethylation,and lead to the loss of m RNA expression,the expression of ER-?gene may occur as a result of silence,cognitive impairment.?3?The risk factors of cognitive dysfunction were considered in the level of homocysteine and estrogen.?4?Methylation of Cp G island in the promoter region of ER-?gene was not related to the decrease of estrogen level.?5?Cognitive impairment in patients with ER alpha gene promoter Cp G island hypermethylation and serum t Hcy concentration was positively related to hyperhomocysteinemia is likely to promote the occurrence of cognitive impairment by interference ER alpha gene methylation and development.
