Objective: The purpose of this study is to understand serum lipid levels and the prevalence of the dyslipidemia in the middle-aged and elderly population of the urban community in Ningxia. To explore the association between blood lipid levels with estrogen levels. To study the association between dyslipidemia and estrogen receptor gene polymorphism.Methods: A cluster sampling method was used to select five communities in the cities of Yin Chuan and Wu Zhong. All meet the inclusion and exclusion criteria. The questionnaire was used in the investgation, and to examine the height, weight, blood pressure and other general health conditions. The blood samples were drawed to test triglycerides(TG),cholesterol(TC),high-density-lipoprotein-cholesterol(HDL-C), low-density lipoprotein cholesterol(LDL-C) and other biochemical indexes, and the serum estradiol(E2) level. The DNA was extracted form whole blood by using Promega DNA abstract Kit. The genotype of ER? gene SNPs(rs2234693?rs9340799)was performed using the Sequenom Mass ARRAY platform. SPSS17.0 software,Plink and Haploview4.2 were used for statistical and genetical analysis.Results1?The lipid levels and the prevalence of dyslipidemia in middle-aged and elderly population in urban community of Ningxia(1) The average of the main components of lipids in the old in ningxia city community : TC(4.84±0.99)mmol/L, TG(1.85±1.05)mmol/L, HDL(2.81±1.66)mmol/L, LDL(3.07±0.79)mmol/L.For ten ages, the levels of HDL and LDL in both male and female of hui ?the levels of TC, TG, HDL, LDL in the male of han ?the levels of TG, HDL levels in the female of han all have the differences between the ages(P < 0.05).(2) The prevalence of dyslipidemia was 38.75%, Whereas ethnic-specific prevalence of dyslipidemia was 40.28% in Han ethnic group and 34.05% in Hui ethnic group(P<0.05),The prevalence of dyslipidemia have no statistically significant difference between male and female of hui ethnic group.But The prevalence of dyslipidemia in male is higher than that in female of hui ethnic group.(P<0.05). The prevalence of high LDL-C and high HDL and hypercholesteremia all have the differences between male and female of hui and han ethnic group.(P<0.05).(3) The prevalence of dyslipidemia decreased with the increasing age in female before 75 age. In the aged 45 to 55 of female,the prevalence of dyslipidemia was lower than that of male,After 55 years old the prevalence rate was increased significantly in the female than the male.2?The association of estrogen and serum lipid levels in middle-aged and elderly population urban community of Ningxia(1)The average level of estrogen in the male was(28.35±20.62)pmol/L, in the female was(26.77±18.82)pmol/L. There was no significantly difference in the male and female. The level of estrogen in the male was increased with the age then falling rapidly after 75 years,there were significantly differences between different age groups(P<0.05). The level of estrogen in the female was decreased with the increased age,there were significantly difference between different age groups(P<0.05).(3)Both male and female, the level of estrogen was significantly lower in the group of high HDL-C than that in the HDL-C normal group(P<0.05). After adjusted by age and BMI, in han ethnic group, as the level of E2 decrease, the level of HDL decrease,and the level of TC increase;In hui ethnic group,as the level of E2 decrease, the level of HDL decrease in the male(P<0.05),but there was no find in female?3?The association of estrogen receptor gene polymorphisms(rs2234693?rs9340799) and serum lipid levels in middle-aged and elderly population urban community of Ningxia.(1) For the SNP rs2234693, total of 1827 DNA samples were genotyped. The frequency of C alleles was39.6%, The frequencies T alleles was 60.4%, The number of CC,CT,TT genotypes was 287?872?668 respectively. For the SNP rs9340799,total of 1800 DNA samples were genotyped.The frequency of G allele was20%, The frequency of A allele was 80%, The number of GG,AG,AA genotypes was 75,571,1154 respectively.(2) SNP rs2234693,the frequency distribution in Ningxia hui population was the same with that in han(p > 0.05). The distribution of each genotype of rs9340799 is the same with that in han(p >0.05).(3)SNP rs2234693,the levels of total cholesterol in the genotype TT group was higher than that in the TC group(P<0.05). The risk of lower HDL-C in subjects with C allele of rs2234693 was 1.27 times higher than that in subjects with T allele(OR=1.27,P<0.05). After adjust by the gender, age, estrogen levels was 1.31 times higher(OR=1.31,P<0.05)in subjects with T allele. ER? gene rs2234693-rs9340799 haplotype T-A group may reduce the risk for lower HDL-C than others.Conclusion The prevalence of dyslipidemia in han ethnic group in middle-aged and elderly in the community of Ningxia was higher than that in hui ethnic group,and that in male is higher than that in female. There were correlations between E2 and HDL?TC in the male subjects(both hui and han ethnic group).There was correlation between the leves of E2 and HDL in the female of han ethnic group. No finding between the levels of E2 and serum lipid in the female of hui ethnic group. The distribution of rs2234693 and rs9340799 SNPs has no ethnic differents. The level of cholesterol in the TT group is higher than that in the TC group. The allele of C may be a risk factor for lower HDL-C level. ER? gene rs2234693-rs9340799 haplotype T-A group may reduce the risk for lower HDL-C than others. There was no correlation between the rs9340799 and the levels of serum lipids.
