Introduction: To investigate the effects of PKC on the aerobic exercise-induced regulation of the functional remodeling of BKCa channels in cerebral artery smooth muscle cell from SHR.Methods: Twenty-four 12-week-old male Wistar–Kyoto rats(WKY,n=24)and Twenty-four 12-week-old male spontaneously hypertensive rat(SHR,n=24),all of them were respectively randomly assigned to sedentary control groups(WKY-SED,n=12;SHR-SED;n=12)and exercise training groups(WKY-EX,n=12;SHR-EX;n=12).The aerobic exercise groups underwent a 12-week motor treadmill training(20m/min,60min/day,5days/week,12weeks).After 12 weeks,the rats were anesthetized by an intraperitoneal injection of sodium pentobarbital solution,the cerebral arteries were removed carefully and cleaned of fat and connective tissue.Acute isolated by enzymatic digestion to obtain a single cerebral artery smooth muscle cell(CASMC).Perforated patch-clamp recording was applied on the freshly separated CASMCs.Results: 1)Before aerobic exercise,compared with the WKY-SED group,SHR-SED group had no significant difference in body weight(P>0.05),HW/BW,SBP were significantly increased(P<0.05),aerobic exercise can significantly improve this change in SHR-EX group(P<0.05),while in WKY-EX group aerobic exercise significantly decreased body weight(P<0.05),and significantly increased HW/BW,SBP(P<0.05);2)PKC can inhibit the amplitude and frequency of STOCs,moreover,the inhibition of hypertension group was significantly higher than that of normal blood pressure group(P<0.05);3)PKC significantly enhanced the amplitude of BKCa whole cell current,furthermore,compared with the WKY groups,PKC had a more significant effect in SHR groups(P<0.05);4)Ang II significantly increased the amplitude of BKCa whole cell current,compared with the WKY groups,Ang II had a more significant effect in SHR groups(P<0.05);5)Compared with the SHR-SED group,aerobic exercise can attenuate the inhibition of PKC on STOCs in SHR-EX(P<0.05),although compared with WKY-SED group,aerobic exercise slightly enhanced the inhibition of PKC on STOCs in WKY-EX,but it did not differ significantly(P>0.05);6)Compared with the SHR-SED group,the enhancement of PKC on BKCa whole cell current was significantly inhibited in SHE-EX(P<0.05),while compared with WKY-SED group,aerobic exercise can significantly enhance the enhancement of PKC in WKY-EX(P<0.05);7)Compared with the SHR-SED group,aerobic exercise can significantly weaken the enhancement of Ang II on BKCa whole cell current in SHR-EX(P<0.05),although compared with WKY-SED group,aerobic exercise slightly enhanced the effect of PKC on BKCa whole cell current in WKY-EX,but it did not differ significantly(P>0.05).Conclusion: In spontaneously hypertensive rats,the enhancement of PKC on BKCa whole cell current is upregulated,however,long-term regular aerobic exercise can significantly suppress this pathological change.
