Objectives:Nine months old male Wistar rats were subjected to HIIT and moderate intensity exercise for 12 weeks,The changes of soleus muscle index,the level of myocyte apoptosis and related apoptotic signals in aging rats were observed,to compare the effects of HIIT and moderate-intensity exercise on the soleus muscle index,myocyte apoptosis and related apoptotic signals in aging rats,so as to provide a theoretical basis for early intervention of HIIT on Sarcopenia.Methods:Eighty male Wistar rats were randomly divided into the basal group?B,n=8?,the quiet control group?C,n=24?,moderate-intensity group?M,n=24?HIIT group?H,n=24?.C group did not exercise,M group were treadmill trained?60%VO2max?,H group were treadmill trained?80%VO2max,40%VO2max high and low intensity alternating?,5 times a week,each time 50min.Rats sacrificed at 1,4,8,12weeks respectively.The soleus muscle index and the expressions of Bcl-2,Bax,Cyt-C,Caspase-3 and PARP-1 in rat were measured.Results:1.Bcl-2,Bax,Cyt-C,Caspase-3,and PARP-1 all have the main effects of exercise mode and time,and the interaction effects of exercise mode and time.2.In the natural aging group,the soleus muscle index was significantly decreased in the12th and 4th week group?p<0.05?;Bcl-2/Bax increased significantly?p<0.05?in the8th and 12th week groups,and Cyt-C increased significantly?p<0.01?in the 8th week;Caspase-3 and PARP decreased significantly in the 8th and 12th week groups.?p<0.01?.3.In the moderate-intensity aging group,the Bcl-2/Bax:12-week group was significantly higher than the 4th week group?p<0.05?.Cyt-C was significantly elevated in the 4th and 12th weeks groups?p<0.01?;Cleaved Caspase-3:peaked at4th week.The 4th week group was significantly elevated?p<0.01?;the 8th week group was significantly higher?p<0.05?.PARP-1:Significant decreased in the12-week group?p<0.05?.4.In the HIIT ageing group,Bcl-2/Bax:Significant decrease in the 4-week group?p<0.05?Cyt-C:Significant increase in the 4th,8th,and 12th week groups?p<0.01?;Caspase-3:4th week Peaked.It was significantly increased in the 4th and 8th weeks groups?p<0.01?,significantly higher in the 12th week group?p<0.05?,and significantly lower in the 8th weeks group than in the 4th week group?p<0.01?.PARP-1:Increases and decreases over time,peaking at 4th weeks.The 4th week group increased significantly?p<0.05?;the 8th and 12th weeks groups showed a significant decrease?p<0.01?.5.4 weeks of exercise intervention,compared with C group,Bcl-2/Bax values in group M decreased significantly?p<0.05?,and group H decreased significantly?p<0.01?.Cyt-C releasing,compared with the C group,M,H group increased significantly?p<0.05?;Cleaved Caspase-3,compared with C group,H group was significantly increased?p<0.01?;PARP-1,compared with M group,H group was significantly increased?p<0.01?.6.8 weeks of exercise intervention,Soleus index increased significantly in group H compared with group C?p<0.05?.Compared with C group,Bcl-2/Bax decreased significantly?p<0.05?.Cyt-C releasing,compared with the group C,M group decreased significantly?p<0.05?,H group increased significantly?p<0.05?;Caspase-3,compared with C group,M,H group were significantly increased?p<0.01?;PARP-1,compared with C group,M group were significantly increased?p<0.01?,H group decreased significantly?p<0.05?.7.12 weeks of exercise intervention,Soleus index was significantly higher in group M than in group C?p<0.05?.Compared with C and M groups,Bcl-2/Bax decreased significantly?P<0.05?.Cyt-C releasing,compared with M group,H group decreased significantly?p<0.05?;Caspase-3,PARP-1,compared with C group,M group was significantly increased?p<0.01?;H group was significantly increased?p<0.05?.Conclusion:1.From 9 to 11 months of age,the soleus muscle index of rats decreased,exercise intervention can effectively increase the soleus muscle index,and the effect of HIIT was superior to moderate intensity exercise at 8 weeks.2.From 9to 11 months of age,the level of apoptosis in rat soleus muscle decreased,exercise intervention can increase the level of Myocyte apoptosis,the effect of HIIT was superior to moderate intensity exercise at 8 weeks.
