| Objective: This study focus on the situation of satellite cells and the level of macrophage inflammatory response in type 2 diabetes after 4 weeks of eccentric exercise,which aim to explore the cause of function change of satellite cells in the condition of type 2 diabetes.Methods:39 male SD rats were divided into four groups: Control(C,n=6),Eccentric exercise(E,n=9),Diabetes(D,n=8),and Diabete-eccentric exercise groups(DE,n=12).E group and DE group were submitted to eccentric exercise on a treadmill down at-5°decline.The position of M1/M2 macrophage and CD68,CD163,MyoD,M-cadherin protein were determined by Immunofluorescence and Western blot.Results:1)This study established a Type 2 diabetesanimal model successfully with 9 weeks of high fat diet feeding after STZ injection.2)After 4 weeks of eccentric exercise,FINS in C group is much lower than E group(P<0.01);HOMA-IR in E group is much lower than C group(P< 0.01),and DE group is lower than D group(P<0.01);ISI in E group is much higher than C group(P<0.01);3)HE results,C group showed no significant difference,E and DE group with varying degrees of inflammatory cell infiltration;4)Western blot results: MyoD protein in E group was higher than C group(P<0.01);Compared with C group,M-cadherin protein in E group was higher(P<0.01);M-cadherin protein in DE group was much lower than E group(P<0.01);CD68 protein in DE group showed significant difference with D and E group(P< 0.01),CD163 protein in E group were much higher than C group(P< 0.01)and DE group were much lower than E group(P< 0.05);5)Immunofluorescence results: CD68 and CD163 protein in E showed significant difference with C group(P<0.01),CD68 protein in DE group showed significant difference with D group(P<0.01),CD163 protein in DE group showed significant difference with E group(P< 0.01).Conclusions:1)4 weeks of eccentric exercise can increase the proliferation and differentiation of muscle satellite cells in normal rats,which showed deeply levels of macrophage inflammatory response;2)4 weeks of eccentric exercise can increase the proliferation of muscle satellite cells in type 2 diabetes rats but with no significant difference in skeletal muscle remodeling,which may be linked with the decrease of M2 macrophage gathering. |
