Objective: To research the effect of exercise and HIF-1? high-expression on the Nrf2 and its target genes in the skeletal muscle of mice.Methods: According to weight,the C57BL/6J mice and HIF-1? high-expression mice were divided into 4 groups randomly.Before the training,the exercise groups mice running 10 min each day for adaptive.In the formal test,the mice did the incremental loading and running,with the initial speed of 10m/min for 10 min.After that,3m/min was increased at every 5min speed,and the slope was 5%,until the mice was exhausted.The mice were sacrificed immediately after the exercise.Enzyme-linked immunoassay(ELISA)determined Nrf2-ARE binding activity in skeletal muscle nuclei.The real-time fluorescence quantitative PCR method was used to determine the mRNA expression levels of the skeletal muscle superoxide dismutase 1(SOD1),superoxide dismutase 2(S0D2),catalase(CAT),and quinone oxidation-reductase(Nqo-1).Western Blot method was used to determine the expression of antioxidant proteins in skeletal muscle HIF-1,Nrf2,SOD1,S0D2,CAT and Nqo-1.The high quality fluorometric method was used to detect the skeletal muscle ROS level in mice.Results:(1)In rest state,compared with wild mice,HIF-1? high-expression mice,skeletal muscle Nrf2 mRNA and protein expression increased significantly,Nrf2-ARE binding activity increased significantly,its downstream target genes SOD1,SOD2,NQO-1,m,RNA expression increased significantly,NQO-1 protein expression increased,skeletal muscle ROS levels dropped significantly.(2)After running,the expression of Nrf2 mRNA in the wild mice was significantly increased,and the combined activity of nrf2-are increased significantly.The expression of skeletal muscle SOD1 and SOD2 mRNA increased,and ROS increased.There was a significant increase in the content of Nrf2 in the skeletal muscle of the HIF-1? high-expression mice movement group.The expression of SOD2 and nqo-1mRNA increased significantly,and the expression of Nqo-1 protein increased significantly,and the ROS showed no significant change.(3)After running,the expression of Nrf2 mRNA and protein expression in HIF-1? high-expression mice was significantly increased compared with that of wild mice,and the expression of Nrf2 nuclear protein and pNrf2 was increased.Skeletal muscle SOD2 and Nqo-1mRNA expression increased,the expression of Nqo-1 protein increased,and skeletal muscle ROS content decreased significantly.Conclusion: 1.Proper HIF-1? high-expression can improve the antioxidant capacity of skeletal muscle in mice,and play a positive role in oxidative stress caused by exercise.2.Exercise can lead to the phosphorylation of Nrf2 and Nrf2 in skeletal muscle of HIF-1? high-expression mice to regulate oxidative stress in the body.
