Purpose: In this paper,we would like to determine the influence of forced exercise on the physiological indexes which include the neurotransmitter in the brain reward pathway and the hormone related to stress in different exercise voluntariness rats,and explore the difference of the influence of forced exercise on motion state and psychophysiological health in different exercise voluntariness rats.Methods: First,sixty SD rats which were six weeks old started voluntary wheel exercise for two days for screening.Used the average of twice of voluntary wheel exercise circles.If the value of one rat is greater than the mean+ standard of voluntary wheel exercise circles,it would be the one of the high exercise voluntariness group(n=12).If the value is less than the mean-standard of voluntary wheel exercise circles,it would be the one of the low exercise voluntariness group(n=12).If the value is between the meanąstandard of voluntary wheel exercise circles,it would be one of the medium exercise voluntariness group(n=36).The research object was the twenty four rats in the high and low exercise voluntariness group.Sort these rats according to the number of voluntary wheel exercise circles level.If the number is odd,the rat belongs to the exercise group,and if ordinal,then goes to the control group.There were divided into four groups: high exercise voluntariness exercise group(HVE),high exercise voluntariness control group(HVC),low exercise voluntariness exercise group(LVE),low exercise voluntariness control group(LVC).The exercise group each week trained five days,and each day trained thirty minutes,training five weeks.The intensity of forced exercise was 75%VO2max which was medium intensity.The control group was just bred.After 24 hours of the exercise intervention,we extracted the serum,the middle brain and the cortex from the blood in a quiet state,and then carried out the biological test.The contents of dopamine(DA),norepinephrine(NE),glutamic acid(Glu),serum 5-hydroxytryptamine(5-HT)and norepinephrine(NE)in the mesencephalon were measured by high performance liquid chromatography-mass spectrometry.The contents of serum corticosterone(CORT)and serum testosterone(T)were measured by enzyme-linked immunosorbent assay.We used two-way analysis of variance to analyze the data.Results: 1.Compared with the LVC group,the DA content in the middle cerebral increased significantly in the LVE group(P<0.01).Compared with the LVE group,the DA content in the HVE group reduced significantly(P<0.01).2.Compared with the LVE group,the content of NE in the HVE group decreased significantly(P<0.05).3.Forced exercise had no significant effect on the content of cortical glutamate in the rats of different exercise voluntariness(P>0.05).4.Forced exercise had no significant effect on the content of serum corticosterone and testosterone in the rats of different exercise voluntariness(P>0.05).5.Forced exercise had no significant effect on the serum serotonin and norepinephrine content in the rats of different exercise voluntariness(P>0.05).Conclusion: 1.Forced exercise can promote the content of catecholamines neurotransmitters in the brain reward pathway,to improve the psychophysiological health of the rats owning different exercise voluntariness,in the meantime,the effect of psychophysiological health improvement of low exercise voluntariness rats is more obvious.2.There is no distinct difference in the effect of forced exercise on the glucocorticoid related to stress of the rats owing different exercise voluntariness.
