Obejective:To investigate the effects of 4 weeks of aerobic exercise mediated apelin on the UCP3 of skeletal muscle in mice.In order to reveal the molecular mechanism by which exercise affects the physiological function of skeletal muscles.Methods:The C57BL/6J wild mice were randomly divided into saline control group?SC?,saline exercise group?SE?,F13A control group?FC?,and F13A exercise group?FE?,with 10 mice in each group.Mice were intraperitoneally injected with F13A?0.2?mol/kg/day?or saline for 4 weeks.Mice in exercise groups underwent 60min/day treadmill running at a speed of 15 m/min with a slope of 5o.After 2 weeks,the maximal oxygen uptake was measured and the running speed was adjusted to 20m/min.The treadmill running continued 4 weeks.The intake and weight of mice were monitored during the period of intervention.The mice were individually housed in a Comprehensive Lab Animal Monitoring System between the 4th week of training to mesure Respiratory Quotien?RQ?and lipid oxidation.Plasma and triceps surae was collected 48 hours after the last exercise.Plasma apelin level was measured by Elisa.The expression of apelin,APJ,AMPK?,p-AMPK?(Thr172),PGC-1?and UCP3 in skeletal muscle were analyzed by Western Blot.Results:1)Compared with saline control group,the expression of apelin of skeletal muscle in F13A control and saline exercise group was significantly increased.2)Compared with F13A control group,the expression of apelin,APJ,AMPK?,p-AMPK?,PGC-1?and UCP3 in F13A exercise group were significantly increased while food intake and lipid oxidation increased significantly,oppositely,the RQ of mice and the plasma content of apelin was markedly decreased.3)Compared with saline exercise group,the RQ of mice in F13A exercise group were significantly decreased.Conclusion:F13A can activate the apelin/APJ system in skeletal muscle under the action of aerobic exercise,increase AMPK activity and up regulate the protein expression of PGC-1?and UCP3,and promote lipid oxidation of skeletal muscle.
