Objective:To explore the effect and mechanism of corn Oligopeptide on improving exercise performance of ICR mice and preventing myocardial injury caused by exhaustive exercise at one time,to provide experimental basis for the application of corn oligopeptide in sports medicine.Methods:40 healthy ICR mice aged 8 weeks were selected.Body weight(30±2)g,divided into 5 groups:NC group,SC group,SL group,SM group,SH group,8 mice in each group.Five groups of mice were given normal saline,0.2 g/kg corn oligopeptide,0.4 g/kg corn oligopeptide and 0.6 g/kg corn oligopeptide by intragastric administration in the morning.Swimming training in the afternoon 45 minutes except the quiet control group,a total of 8 weeks,the last day of the experiment to determine 5%of the body weight of mice in five groups exhausted swimming experiment,record exhausted swimming time.Blood lactate levels were measured before,immediately after,5 min after,15 min after and 30 min after exhaustive exercise.The mice were executed the next day after exhaustion test,collect blood,heart,samples,using enzyme-linked immunosorbent method to detect serum CK,CK-MB,SOD,MDA content,western blot detecting autophagy and apoptosis in myocardial tissue and the expression of antioxidant related proteins and immunofluorescence detection of apoptosis,to explore the effect of exercise combined with corn oligopeptide intervention on myocardial protection.Results:1)Compared with the SC group,the weight of mice in the SH group increased significantly(p<0.05).There was no significant change in SL and SM groups.2)The swimming time of exhaustion showed an obvious increasing trend with the increase of corn oligopeptide concentration,but there was no significant difference among the groups.3)Blood lactic acid level increased after exhaustion,with the most obvious increase in SC group,SL,SM,SH group were basically lower than the NC group,and recovered to the normal level after 30 minutes of exhaustion,the fastest recovery in SL group.4)Compared with the NC group,serum CK of mice in SL group decreased significantly(p<0.01),serum CK of mice in SM group and SH group decreased significantly(p<0.05),and serum ck-mb content of mice in SL group decreased significantly(p<0.05).Although there was no significant difference in other groups,there was a downward trend.Compared with SC group,serum CK in SL group was significantly decreased(p<0.01).5)TUNEL results showed that,compared with the NC group,the apoptosis rate of myocardial cells in the SL group decreased significantly(p<0.05),the apoptosis rate of myocardial cells in the SH group decreased significantly(p<0.01),and the apoptosis rate of myocardial cells in the SH group decreased significantly(p<0.05)6)Based on the type of myocardial apoptosis related proteins table testing,compared with NC group,the SL group mice myocardial volume of the expression of Bcl02 significantly increased(p<0.05),SM?SH group Bax expression in myocardium in mice significantly lower,compared with the SC group,SL group mice myocardial volume of the expression of Bcl-2 significantly increased(p<0.05).7)Based on the type autophagy related protein expression were detected,compared with NC group,SC group had elevated levels of autophagy,Atg7,LC3 expression quantity increased significantly(p<0.05),movement joint SL,SM,SH mice after drug intervention myocardial autophagy level increased significantly,Atg7,Beclin 1,p62,LC3 expression quantity increased significantly(p<0.05).8)Compared with the NC group,the expression of Nrf2 in the myocardium of the SL and SH groups was significantly increased(p<0.05),and the expression of Keapl in the myocardium of the SL and SH groups was significantly increased(p<0.05).Compared with SC group,the expression of Keapl in the myocardium of SL group was significantly increased(p<0.05).Compared with the NC group,serum SOD in the SL group increased significantly(p<0.05),MDA content in the SC group decreased significantly(p<0.05),and MDA content in the SL and SM groups decreased significantly(p<0.01)Conclusion:1)Exercise combined with corn oligopeptide group can effectively prolong exhaustive exercise time and improve the aerobic exercise ability of mice.High dose group has better effect.2)Exercise combined with corn oligopeptide group can effectively reduce CK and CK-MB in serum of mice,activate autophagy,reduce apoptosis,improve antioxidant capacity,and have a certain protective effect on myocardium.The effect of low dose group is better.3)The myocardial protective effect of exercise combined with corn oligopeptide group was better than that of swimming control group.
