Objective: through the different load swimming experiment, with the natural aging of16rats as the research object, explore the Bcl-2swimming exercise on myocardialcells, Bax protein expression changes and HIF-1, attempt to summarize differentload swimming exercise and myocardial cells the Bcl-2, Bax between control groupand the relationship of the expression of HIF-1, and to further explore aging rats inswimming exercise intervention effect and the mechanism of myocardial cellapoptosis, to exercise intervention aging in the search for a better, more reasonableand more effective ways to promote exercise, providing theoretical basis for thesuitable set of exercise intensity and the basis.Methods:48aged rats with natural aging male health Wister random points (eachgroup of12): the control group (conventional breeding), big load group H C (60min/d), medium load group M (30min/d) and small load group S (15min/d) for8weeks, training six days a week (Sunday) rest of swimming exercise, stay after theexperiment immediately to dissect for HE staining in the rat myocardial cellmorphology observed between groups of experimental animals, the BCL-2immunohistochemical detection of myocardial cells, Bax and HIF-1proteinexpression, analysis of the indexes in different load swimming exercise and differenceand correlation between the control group.Results: the weight in weight before and after the experiment group C there was nosignificant difference (P>0.05), group H after test weight shows have significantdifference (P <0.05), group M after test before the experiment, but there was nosignificant difference, the group S weight increase obviously before and after theexperiment, and have significant difference (P <0.05); Group C myocardial fibersarranged disorderly, gap width, interstitial, more obvious hypertrophy, myocardialfibers of fracture H group, capillary hyperemia, blank area, the largest necrosis; Mgroup of cardiac muscle fiber fracture, mild hyperemia, no necrosis, S group of myocardial fibers hypertrophy, towards normal, uniform color, no fault, no congestion,no necrosis occurs.Each group the Bcl-2protein expression in myocardial cells as: S group> M> groupC> group H, S group of aging rats myocardial Bcl-2expression and C group (P <0.01) difference was very significant, H and C group (P <0.05) differencesignificantly, M group there was no significant difference (P>0.05); S and M, Hgroup, group S the Bcl-2expression in myocardial cells and M group (P <0.01), andgroup H (P <0.01) were very significant differences; Bax protein expression inmyocardial cells as group H> C> M> S, group C S myocardial Bax expression andC group (P <0.01) difference was very significant, H and C group (P <0.05)difference significantly, M group there was no significant difference (P>0.05); S andM, H group, group S aging Bax expression in rat myocardial cells and M group (P <0.01), and group H (P <0.01) were very significant differences, myocardial cell alphaHIF-1protein expression situation is: the group S> C> M> H, the group Smyocardial cell HIF-1alpha expression and group C (P <0.05) differencesignificantly, H and C group (P <0.01) difference was very significant, M and Cgroup there was no significant difference (P>0.05); S and M, H group, group S agingHIF-1alpha expression in rat myocardial cells and M group (P <0.01), and group H(P <0.01) were very significant differences.Conclusion: the aged rats with different load swimming exercise on myocardial cellthe Bcl-2, Bax and express different HIF-1. With the increase of exercise load, theBcl-2in the aging rat myocardial protein positive rate showed a trend of decreasingto reduce; Bax protein positive rate increasing trend; HIF-1alpha protein positiverate will decrease trend to decrease. Show that the proper swimming group (S) help toincrease the Bcl-2expression, Bax protein expression is reduced, HIF-1a higherexpression, thereby inhibiting myocardial apoptosis occurred; Big load movement orinappropriate group (H, M) for the Bcl-2protein expression is reduced, theexpression of Bax protein increases, reduced HIF-1a expression, thus promote theoccurrence of myocardial cell apoptosis. Tip: the Bcl-2, Bax protein expressionchanges is one of exercise intervention regulatory mechanism of myocardial cell apoptosis, and in aged rats with different load swimming exercise on HIF-1alphaexpression in myocardial cells consistent with the trend of the Bcl-2proteinexpression, were positively correlated. And coordinated the Bcl HIF-1-2expressionof myocardial cell protection mechanism needs further research.
