| ObjectiveTo study the effects of hypoxia exposure combined with increasing load exercise on body weight,white fat,brown fat and blood lipid in obese mice and the changes of UCP1 protein expression in the process.To explore the effect of increasing load hypoxia training on lipid metabolism in mice and its possible mechanism.MethodsSeventy 3-week-old male C57BL6 mice were randomly fed with 12 normal diets and 58 high-fat diets.At the end of four weeks of feeding,the average weight gain of each high-fat-diet mouse was greater than 20% of the average weight of each ordinary-diet mouse as the obesity standard.The obese mice were screened out after adaptive training and 24 mice were selected.They were randomly divided into obesity control group(OC group),obesity low living low training group(OLT group),obesity high living high training group(OHH group)and obesity high living low training group(OHL group).Six mice were randomly selected as general diet control group(N group).During the experiment,the mice in the training group were trained on the level animal treadmill,and the mice in the training group were trained once a day for 6 days a week for a total of 4 weeks.The training program under normoxic condition was as follows: week 1: 18 m / min,continuous exercise 30 min,week 2: 18 m / min,continuous exercise 40 min,week 3: 20 m / min,continuous exercise 40 min;Week 4: 20 m / min,continuous exercise 50 min,training program under hypoxia: 16 m / min,30 min;week 2: 16 m / min,continuous exercise 40 min;week 3: 18 m / min,continuous exercise 40 min;Week 4: 18 m/min,continuousexercise 50 min.The hypoxia environment used in the experiment is a simulated atmospheric hypoxia environment at an altitude of 3500 m.Daily intake was recorded during the experiment,and mice were weighed weekly.At the end of experiment 4,after the last exercise rest for 24 hours in the training group,the blood samples were collected from the abdominal aorta to separate the serum and the blood lipids were measured,and the perirenal fat,epididymal fat and brown fat were taken from the mice.The expression level of UCP1 protein in brown adipose tissue was measured by.Western blot method in gastrocnemius muscle.Result1.After four weeks of high-fat feeding,the weight of obese mice was higher than that of mice on normal diet,and the difference was very significant(p < 0.01).Triglycerides(TG),total cholesterol(TC)and LDL-C of the obese mice were significantly higher than those of the normal diet mice(p < 0.05),while HDL-C of the obese mice was lower than that of the normal diet mice(p < 0.05).2.At the end of the fourth week of the experiment,the weight of mice in both the OHH group and the OHL group was significantly lower than that in the OC group(p < 0.05),and there was no significant difference between the OHH group and the N group and the OLT group,and there was no significant difference between the two groups.Compared with the weight before the experiment,the weight of mice in both the OHH group and the OHL group at the end of the fourth week was significantly lower than that before the experiment(p < 0.05),and the weight drop of the two groups was significantly higher than that of the other groups(p < 0.05),but there was no significant difference between the two groups.3.At the end of four weeks,the level of TC,TG in OHH group and OHL group was significantly lower than that in OC group(p < 0.05).There was no significant difference between OLT group and N group,but there was no significant difference between the two groups.The level of LDL-C in OHH group and OHL group was significantly lower than that in OC group,and there was no significant differencebetween OLT group and N group,and there was no significant difference between the two groups.The level of HDL-C in OHH group was significantly higher than that in OC group,but there was no significant difference between OC group and other groups.There was no significant difference in HDL-C level between OHL group and other groups.There was no significant difference between OHH group and OHL group.4.At the end of four weeks,the fat body ratio in OHH group was significantly lower than that in other groups(p < 0.05).The fat body ratio of OHL mice was significantly lower than that of OC group(p < 0.05),and there was no significant difference between OHH group and OLT group and N group.The perirenal fat of OHH group was significantly lower than that of other groups(p < 0.05),and the epididymal fat of OHH group was significantly lower than that of OC group and OLT group.There was no significant difference between OHL group and N group.The peri-renal fat and epididymal fat in OHL group were significantly lower than those in OC group(p < 0.05),but there was no significant difference between OLT group and N group.5.At the end of the fourth week of the experiment,the relative amount of brown fat of mice in the OHH group and the OHL group was significantly higher than that of the other groups(p < 0.05),and there was no significant difference between the two groups.The expression level of UCP1 in the brown adipose tissue of mice in the OHH group and OHL group was significantly higher than that in the N group and the OC group(p < 0.05),and there was no significant difference between the two groups and the OLT group,while there was no significant difference between the two groups.6.During the experiment,the food intake of mice in the OHH group and OHL group was relatively low,in which the food intake of mice in the OHH group was79.4% of that in the OC group.The food intake of OHL group was 84.1% of that of OC group.Conclusion1.The high-fat Diet was adopted to feed C57BL6 mice at 3 weeks of age for 4weeks,and 20% of the average body weight of the mice with normal Diet was taken as the obesity standard,which could effectively establish the obesity mouse model.2.Both high and low training can effectively reduce the body weight of obese mice,and have a good effect on the level of blood lipid in obese mice,to a certain extent,the effect is better under the mode of high living and high training.3.High and low training could keep the perirenal,epididymal fat content,fat /body ratio,and high muscle content in mice.To a certain extent,the effect of high training and fat reduction is more significant.4.High living,high training and high and low training could increase the relative amount of brown fat and the expression of UCP1 protein,and the higher expression of UCP1 protein could increase the level of energy metabolism in mice,which may be related to the weight loss of mice. |
