Effects Of Hypoxia Combined With Endurance Exercise On The Expression Of Mitochondrial Autophagy-related Factors In Skeletal Muscle Of Obese Rats

Objective:In this experiment,we established a hypoxic endurance exercise model of obese rats to study the effects of three hypoxic concentrations(11.3%,13.3%,16.3% O2concentration)combined with endurance exercise on skeletal muscle mitochondrial autophagy-related factors in obese rats.The expressions of mitochondrial autophagy-related factors FUNDC1,BNIP3,PINK1 and Parkin were detected by Western blot and RT-PCR.The effects of hypoxia combined with endurance exercise on the expression of mitochondrial autophagy-related factors in rat skeletal muscle were investigated.And provide a theoretical reference for the prevention of mitochondria-related diseases.Mehhods:A rat model of nutritional obesity was constructed and randomly divided into eight groups: normal oxygen quiet group(group A),normoxia exercise group(AE group),16.3% hypoxic quiet group(group B),and 16.3% hypoxia.Exercise group(BE group),13.3% hypoxic quiet group(C group),13.3% hypoxic exercise group(CE group),11.3% hypoxic quiet group(D group),11.3% hypoxic exercise group(DE group),10 in each group.The training program of the formal experimental rats was as follows:the exercise group rats used moderate-intensity endurance exercise with a slope of 0°and a running speed of 20m/min,and the exercise time was 40min;the exercise frequency was once a week(Monday to Friday)for 8 weeks.After the last training,fasting for 24 h,the rats were sacrificed and sampled.The biochemical indicators were used to measure blood glucose and blood lipids.The expression levels of mitochondrial autophagy-related factors FUNDC1,BNIP3,PINK1 and Parkin were measured by Western blot.The bones were determined by RT-PCR.Expression of muscle mitochondrial autophagy-related factors FUNDC1,BNIP3,PINK1,Parkin mRNA.Result:1.Nutritional obesity rat model: After 8 weeks of modeling,the body weight,Lee's index,BG,TC,TG,LDL-C concentrations in the high-fat diet group were significantly increased(P<0.05);2.Changes in morphological parameters of rats: Compared with rats in group A,the body weight of rats in AE group,E group and D group and the Lee's index of rats in AE group were significantly decreased(P<0.05);Compared with the AE group,the body weight of the rats in the B group and the Lee's index of the rats in the B group and the C group were significantly lower(P<0.05).3.Western Blot results showed that the expression of BNIP3 protein in AE rats was significantly higher than that in group A after 8 weeks of hypoxia combined with endurance training(P<0.05).Compared with group B,the expression of BNIP3 protein in FUNDC1 protein of group BE and group D was significantly increased(P<0.05).Compared with the BE group,the expression of Parkin and PINK1 protein in the DE group was significantly increased(P<0.05).Compared with the CE group,the expression of FUNDC1 protein in the DE group was significantly increased(P<0.05).4.Real-time PCR results showed that Parkin,PINK1 mRNA in Parkin,PINK1,FUNDC1,BNIP3 and C rats in group AE and group D compared with group A after eight weeks of hypoxic combined endurance training.The expression was significantly increased(P<0.05);Compared with the AE group,the expression of Parkin,FUNDC1 and BNIP3 mRNA in the DE group was significantly increased(P<0.05).Compared with group B,the expressions of PINK1,Parkin,FUNDC1 and BNIP3 mRNA in rats in group C of PINK1,FUNDC1 and D groups were significantly increased(P<0.05).Compared with the BE group,the expression of PINK1 mRNA in the PINK1,Parkin,FUNDC1,BNIP3 and CE rats in the DE group was significantly increased(P<0.05).Compared with group C,the expression of PINK1 and FUNDC1 mRNA in PINK1 and D rats of CE group was significantly increased(P<0.05).Compared with CE group,the expressions of Parkin,PINK1,FUNDC1 and BNIP3 mRNA in DE group were significantly increased(P<0.05).Compared with group D,the expressions of Parkin,PINK1,FUNDC1 and BNIP3 mRNA in DE group were significantly increased(P<0.05).Conclusion:1 Three different oxygen concentrations and combined endurance exercise can effectively reduce the body weight of obese rats.Under the action of hypoxia combined with endurance exercise,the weight loss of rats is more significant,which can effectively reduce the obesity of rats.2 hypoxia combined with endurance exercise can induce mitochondrial autophagy.The expression of mitochondrial autophagy-related factors increased with the decrease of oxygen concentration.Under the intervention of hypoxia endurance exercise,the expression of mitochondrial autophagy-related factors was similar.3 Hypoxia combined with endurance exercise can up-regulate the expression of FUNDC1,BNIP3,PINK1,Parkin factors in skeletal muscle cells to enhance the level of autophagy in mitochondria.
