Objective: This research adopts to the ovariectomized rat model of osteoporosis in postmenopausal women,by measuring the rat body weight,body composition,bone microstructureand,the expression of fat genes in peripheral blood mononuclear cells,exploring the relationship between the changes of the fat gene in peripheral blood mononuclear cells and bone in ovarian rats after moderate-intensity treadmill exercise.In this way,the molecular biological mechanism which exercise uses to retard bone loss is revealed and studied,providing a theoretical foundation for preventting postmenopausal osteoporosis.Methodology: In this study,Sixty 3-month-old female Sprague-Dawle rats are randomly assigned to the following three groups: ham-operated(20 rats)(SHAM),ovariectomy(20 rats)(OVX),and ovariectomy with exercise(20 rats)(OVX+EX)for17 weeks of experimental treatment.One week after the operation,the ovariectomized rats with exercise will take one-week-long adaptable training first,and then followed by treadmill exercise training at a speed of 20m/min and a slope of 5 degrees for 60 minutes,5 times a week for 17 weeks.After the experiment,the rats' weight will be measured,and the body composition will be detected by dual-energy X-ray densitometry.Besides and the femoral bone microstructure will be detected by Micro-CT.Finally,the peripheral blood mononuclear cell(PBMC)will be extracted from blood and the genetic expression in PBMC of LPL,ALOX15 and FNDC5 will be detected by qRT-PCR.Results:(1)Compared with SHAM group,the body weight,total body fat percentage,and percentage of fat in upper limbs,lower limbs and trunkare significantly higher of rats in the OVX group;And at the same time,the total body muscle content and percentage of muscle in upper limbs,lower limbs and trunk are very significantly lower.Compared with the OVX group,the weight of rats in the OVX+EX group is significantly reduced,the fat percentage of lower limbs and trunk is very significantly reduced,the muscle content of the whole body is very significantly increased,the muscle percentage of upper limbs and trunk is very significantly increased,and the muscle percentage of lower limbs is very significantly increased.(2)Compared with the SHAM group,the trabecular microstructure of the distal femoral bone in the OVX group is seriously damaged,with sparse trabecular bone and a large gap between trabeculae.Compared with OVX group,the number of trabeculae in OVX+EX group is increased and the bone continuity increases.(3)Compared with SHAM group,LPL and ALOX15-mRNA levels of peripheral blood mononuclear cells in OVX group are significantly reduced.Compared with OVX group,LPL,FNDC5 and ALOX15-mRNA levels of peripheral blood mononuclear cells in OVX+EX group are significantly increased.Conclusion: Moderate intensity treadmill exercise can reduce the weight and fat content of ovariectomized rats,slow down the decline of muscle content,and improve the bone microstructure of femoral cancellous bone,indicating that moderate intensity treadmill exercise has a preventive effect on the occurrence and development ofosteoporosis.At the same time,it also can promote the expression of peripheral blood mononuclear cells in the fat genes,indicating that LPL,ALOX15 and FNDC5 gene expression in peripheral blood mononuclear cells can be used as potential blood biomarkers for improving osteoporosis by sports.As a result,the expression of these genes provides experimental evidence for the future study of the molecular mechanism of peripheral blood mononuclear cells in the prevention and treatment of postmenopausal osteoporosis.
