Objective:Along with the social population ages, postmenopausal osteoporosis was also to be increased year by year then became a common disease, which seriously affect the human health development. Therefore, the prevention and treatment of postmenopausal osteoporosis has become an important medical problem. In order to investigate the prevention and treatment mechanism of exercise on postmenopausal osteoporosis, this paper observed the effects of the moderate-intensity-exercise on the protein expression of VEGF in bone tissues of ovariectomized (OVX) rats by imitating the condition of postmenopausal osteoporosis by OVX rat model.Methods:Sixty health female SD rats were randomly divided into following tow groups by body weight:sham-operation (Sham, n=20), ovariectomized (OVX, n=40). One week after the operation, the OVX group was randomly divided into tow groups by body weight:sedentary OVX (OVX, n=20) and exercised OVX (EX, n=20). After then, the EX group was trained for 45min at a speed of 18 m/min with 5°lope four times per week for 17 weeks. At the end of experiment, weighed body weight and uterus weight; detected the serum level of E2; detected the femoral and L3 bone mineral density (BMD) and bone mineral content (BMC) by dual-energy X-ray absiorptiometry; detected the femoral and L4 bone biomechanical index; detected the protein expression of VEGF in femoral and L5 by Western blotting; detected the protein location of VEGF in femoral and L5 by immunohistochemistry.Results:(1)The body weight of OVX was significantly higher than that of Sham, but that of EX was slightly lower than OVX with no statistical significance;(2) the uterus weight of OVX was significantly lower than that of Sham, but that of EX was slightly higher than OVX with no statistical significance; (3)the serum level of E2 of OVX was significantly lower than that of Sham and EX With significant difference; (4) the BMD and BMC of distal femur and L3 in OVX were significantly lower than that in Sham and EX, but the proximal and middle of femoral were no significant difference with the other two groups;(5) the maximum load of femur and L4 in OVX was significantly lower than that in Sham and EX; (6) Immunoblotting showed that the protein expression of VEGF of femur and L2 in OVX was significantly decreased compared with Sham; After treated with treadmill exercise, the protein expression of VEGF in EX was significantly increased compared with OVX; (7) Immunohistochemistry showed:compared with Sham, in OVX, the cartilage cells of femoral and the stromal cells on trabeculae of vertebrae were negative expression, the osteoblasts and the osteoclast were weak positive expression; after the sports training, the cartilage cells of femural and the stromal cells on trabeculae of vertebrae were weak positive expression, the osteoblasts and the osteoclast were positive expression.Conclusion:Moderate-intensity treadmill exercise can increase the serum level of E2; prevent the bone mass and the bone biomechanical characteristics of vertebrae and femoral, increase the protein expression of VEGF of femur and lumbar vertebrae in OVX.
