Hericium erinaceus is a kind of precious food and medicine fungi,which has very important biological activity.At present,the development and use of Hericium erinaceus mainly concentrated in the polysaccharides which is separated from Hericium erinaceus fruit body.As raw materials,in this article with Hericium mycelium which was fermented from Hericium erinaceus researchrd the solvent reflux extraction process parameters optimization,the initial structure and biological activity in vitro of Erinacine which was from ethanol extraction active substances.?1?Fermentation of Hericium mycelium was pretreated with enzymatic and acid hydrolysis and then optimized by the method of ethyl ethanol reflux extraction through mono-factorial experiment and response surface methodology?RSM?.The pretreatment with enzymatic and acid hydrolysis can significantly increase the extraction yield of ethanol extraction active substances from Hericium mycelia material.The optimum process parameters for extraction process were obtained:ethanol concent ration 65%,liquid material ratio 1?36 g/mL,extracting temperature63?and extracting time 92 min.The yield of ethanol extraction active substances from Hericium mycelium was increased to 3.39%.?2?The ethanol extraction active substances from Hericium mycelium was extractied by petroleum ether,then was extractied by ethyl acetate.The ethyl acetate extraction which was filtered by 0.45?m organic membrane was analyzed by GC-MS,the result showed that 67.25%of the extracttion are terpenoids and steroids.?3?By employing silica gel column chromatography and semipreparative high performance liquid chromatography,A kind of diterpenoid compounds Fr-3-1 was isolated from fermentation of Hericium mycelium and the purity reached 94.59%by TLC and HPLC determination.On the basis of UV,IR,HPLC-MS and 1H NMR spectral data analysis and related literature,the structure of Fr-3-1 was identified as Erinacine A.?4?The antioxidant activities of Fr-3-1 which was isolated from fermentation of Hericium mycelium crude was evaluated by in vitro assays including superoxide action,DPPH and ABTS.The results showed,in the ranges of the experiment,there was a linear relationship between the reducing power and concentration of Fr-3-1,the EC50 of superoxide action,DPPH and ABTS were 0.93,1.21,0.40 mg/mL,respectively.?5?The inhibiting effects of Fr-3-1 against the growths of bacteria in vitro was studied.The result showed,Fr-3-1 only for Staphylococcus aureus showed good antibacterial activity,and no obvious resistance on Escherichiacoli and Yeast.?6?The inhibiting effects of Fr-3-1 against the growths of human gastric cancer cells?MGC-523?activity in vitro was studied.The IC500 of inhibiting the growths of MGC-523 cells was 453.14?g/mL.?7?The promote on Radial glial cells of rats?Ra?secretion of nerve growth factor?NGF plays?in vitro was aslo studied.The promote of Fr-3-1 on Radial glial cells of rats?Ra?secretion of nerve growth factor?NGF plays?in vitro was better than adrenaline.On the concentration of 100 mu g/mL,the promote of Fr-3-1 for the secretion of radial nerve cells of rats nerve growth factor concentration was 46.63pg/mL,far higher than the same concentration of adrenaline stimulate secretion of nerve growth factor concentration 37.77 pg/mL.
