Hericium erinaceus is a long-established edible and medicinal fungus.It is rich in a variety of physiologically active substances and also has a variety of pharmacological effects.So it has great applications in drug development.When Hericium mycelium is used as the raw material for the development of gastric medicine products,the main metabolite like ergosterol in Hericium mycelium has a certain content requirement.In order to solve this practical problem of low ergosterol content in Hericium mycelium,the high-yield fermentation conditions and extraction process of ergosterol from Hericium erinaceus were studied.An HPLC method for the determination of ergosterol in the mycelium of Hericium erinaceus was established by HPLC chromatography analysis.The method showed good precision,repeatability,and stability,and the sample recovery rate is high,which indicating that the method is reasonable and feasible.Liquid fermentation was used to prepare mycelium of Hericium erinaceus with high-yield ergosterol.Through the seletion of strains,the Hegong strain was identified as the target strain,and soybean meal was selected as the basic inducer of fermentation,and then the best carbon source,nitrogen source,inorganic salt,and inducer for high-production of ergosterol from Hericium erinaceus were seleted through single-factor experiments.Finally,through Box-Behnken experiment optimizatin,the optimal fermentation conditions for the high-yield of ergosterol from Hericium mycelium was determined to be: 3.3% glucose,0.3%peptone,0.15% MgSO4,3.2% soybean meal powder,and the maximum ergosterol yield was5.7770 mg/100 m L.The best extractant,material-liquid ratio,ultrasonic time,ultrasonic temperature,and ultrasonic power for the ergosterol extraction from Hericium mycelium were determined through single-factor experiments.Then through the Box-Behnken test,the best extraction conditions were as follows: analytical grade methanol as extractant,ultrasonic temperature80 ?,material-liquid ratio 1:29,ultrasonic time 97 min,ultrasonic power 180 W.Under these conditions,the extraction yield of ergosterol by Hericium erinaceus is 2.3741 mg/g.
