China is the hometown of tea,the costom of making and drinking tea lasts thousands of history,there are six kinds of tea,namely green tea,black tea,oolong tea,white tea,yellow tea and black tea.Tea contains a variety of active ingredients such as tea polyphenols,tea polysaccharides,theanine,caffeine,dietary fiber,tea pigments,vitamins,minerals and so on,which have a wide range ofphysiological effects.Besides,it is good for healthy and has high nutritional and medicinal value.The use of green tea is the earliest,but also the largest amount of tea,the current green tea generally has three kinds of usage:the way of immersion with boiling water,mainly drinking tea way;active ingredients by solvent extraction and purification are processed into instant tea or health tea;making matcha by ultra-fine grinding,which used in cakes,beverages,candy and other food as additives.However,the way of immersion with boiling water can not be fully dissolved ingredients,utilization is very low,resulting in a great waste of tea resources,on the other hand,hot drink for a long time is also easy to cause some damage to the esophagus;the main active ingredients such as tea polyphenols and chlorophyll in the green tea under the light and air are easy to oxidize,degrade,polymerize,epimerization,and the stability is poor by solvent extraction,purification and other means,what's more,the production process cumbersome,costly,environmental pollution;the use of matcha improves efficiency of green tea,but in the preparation process can easily lead to loss of catechins and other untable substances.The purpose of this project is to develop green tea preparations which are different from trational tea products,take conveniently and have good quality.To promote the full use of green tea nutrients,and improve their dissolution efficiency,solubility,enhance stability;to prompt a variety of green tea ingredients to synergie in the body and increase the bio-utilization of green tea;to provide new ways and ideas for the development and utilization of tea new products by ultra-fine grinding and cyclodextrin inclusion technology.The content of water-soluble tea polyphenols in green tea was determined by UV-Vis spectrophotometry,and the method was validated.Besides,the content of tea polyphenols in green tea was determined according to GB/T8313-2008 method.The possibility of formation of supramolecule of green tea active ingredients and cyclodextrin was studied by UV spectroscopy and calculated binding constants;preparation of green tea/cyclodextrin ultrafine powder(abbreviated as ultrafine powder)was compared by dry and wet grinding,the ultra fine powder by dry method was explored to form the feasibility of supramolecule of the active ingredients and cyclodextrin;The amount of tea polyphenols dissolved in water and cost-effective as indicators to determine the best inclusion ratio;optimum process conditions of dry ball mill was determined through orthogonal test screening;The superfine powder was characterized by DTA,UV-vis DRS and DR-FTIR.The solubilization and stability of the active components were determined by UV-vis,green tea/cyclodextrin ultrafine powder was prepared by dry ball mill.The prescrption and process of the green tea tablets were screened by single factor experiment.And the stability was determined by influencing factor test;the differences were compared between green tea leaves and micro powder tablets by UV-vis and HPLC.The results showed that the active components of green tea could form supramolecules with cyclodextrin,and ?-CD was better than ?-CD;?-CD was used as inclusion material,and the optimum inclusion mass ratio was green tea and ?-CD-1:1;the milling process conditions were 600 r/min,the milling time was 15 min,the mass ratio of the ball to the material was 10:1;DTA,UV-vis DRS,DR-FTIR showed the supramolecular body of green tea and ?-CD has been formed;ultrafine powder makes tea polyphenols solubilize about 2 times and improve the stability,tea polysaccharide solubilization 3 to 4 times;single factor screening of green tea tablets final prescription:69%green tea/?-CD ultrafine powder,20%MCC,10%super sodium carboxymethyl starch,a small amount of 75%ethanol-aqueous solution,1%magnesium stearate,tablet pressure 4?6Kg,total sheet weight 0.65g;Self-made green tea micro-powder chip stability is good in the high temperature,more sensitive to light and humidity by influencing factors test;both UV-vis and HPLC showed that the amount of dissolution of the active ingredients of green tea tablets were significantly higher than that of tea leaves and dissolution faster.In summary,the green tea tablets by the use of ultra-fine grinding and inclusion technology provide new methods and ideas for better development and utilization of tea resources.
