Skin lesions and injuries can increase the risk of pathogen infections.Developing efficacious wound dressings could effectively prevent bacterial infection and accelerate wound healing.Due to its good biological properties,antibacterial hydrogels have received extensive attention as a novel wound dressing.Antibacterial hydrogels loaded with non-antibiotics have made encouraging progress in this regard.With the rapid development of science and technology,personal requirments for wound dressings are getting higher and higher.For the treatment of wounds,it is not limited to simple hemostasis,but higher and more requirements.In this paper,chitosan was used as raw material to prepare physicochemical double-crosslinking hydrogel(noted as DR-CCH)by UV initiation crosslinking technology.The basic properties and of DR-CCH were analyzed.The biological properties and applicability of DR-CCH were tested by antimicrobial test and cytotoxicity test.The promotion of DR-CCH as wound dressing was tested by systematic animal experiments.Results are as follows.1.The water-soluble carboxylated chitosan(CC)is used as a water-soluble macromolecular matrix.Then a physicochemical double crosslinked DR-CCH was prepared under UV irradiation with diazo resin,containing zinc ions and cationic diazonium group,and CC.The double crosslinked structure of the DR-CCH is more stable than the physical crosslinked structure of the H+-CC hydrogel and the Zn2+-CC hydrogel.DR-CCHs have a homogeneous porous polymer network structure,which would permit the loaded zinc ion to release into wound tissue or bacteria.2.This composite DR-CCH could inhibit and kill E.coli and S.aureus and show excellent antibacterial properties.The DR-CCH had the best cell viability and then was selected as dressing for the wound healing of the full-thick skin in vivo.The above results clearly demonstrated that DR-CCH could effectively accelerate wound healing in vivo.The DR-CCHs were expected to develop wound dressings containing trace amounts of zinc for wound treatmentIn the above basic research,a polyethylene glycol(PEG)-based double-layer hydrogel was successfully prepared.In the course of the research,PEG was used as the raw material,and the quaternary ammonium salt was used as the antibacterial agent.Then a PEG-based double-layer hydrogel was prepared by the reaction of generating a double bond between the aldehyde group and the cyano group.The above results inspired us to develop dressings for wound treatment in the following research and clinical utilization.
