The experiment was carried out in the Horticulture Laboratory of Agricultural College of Heilongjiang Bayi Agricultural University in 2018.Na Cl,Na2SO4,Na HCO3 and Na2CO3 were weighed in molar ratios of 1:1:0:0,1:2:1:0,1:9:9:1,1:1:1:1,9:1:1:1:9 respectively to prepare the treatment solution,which was divided into five treatment groups.Each group was treated with different salt concentration.Through the study of seed germination and physiological index changes of different varieties of Isatidis Radix under different salinity-alkali stress,the response rules of Isatidis Radix seeds to salinity-alkali stress at the initial stage of seed germination and seedling growth were summarized.The salt tolerance of different varieties of Isatidis Radix was determined.Regional selection and cultivation provide scientific basis in Daqing area.The primary salinity of soil in Daqing area is relatively high.Combining the improvement and utilization of saline-alkali soil with the planting and production of Isatidis Radix can not only alleviate the harm of soil salinization to the ecological environment,but also produce certain social and economic benefits.The results are as follows:In the germination stage of seeds: under low salinity stress(20 mmol/L p H 7.30),the germination rates o f Bozhou Xiaoye,Anguo-1,Anguo-2 and Anguo-3 were higher than those of the control,and the germinatio n rate of Bozhou Daye was lower than that of the control;under the salinity of 20 mmol/L p H 7.96,the g ermination rate of Bozhou Xiaoye was 86.67%,which was higher than the control of 85.56%;Under the con ditions of 7.30,the germination rates of Anguo-1 were 78.89% and 82.22% respectively,which were higher t han the control of 77.78%.According to the comprehensive comparison of germination rate,germination potential,germination index and vigor index among five varieties,it was found that Bozhou Xiaoye had the best salt-alkali tolerance,followed by Anguo-1,and Bozhou Daye was the most sensitive variety to salt-alkali stress.In the seedling stage: when the p H was 8.64,the chlorophyll content of Isatidis Radix of five cultivars all increased firstly and then decreased with the increase of salt concentration.Compared with the control,the increase of Bozhou Xiaoye was the largest and that of Bozhou Daye was the smallest.The soluble protein content of Bozhou Daye continued to decrease,while the soluble protein content of Bozhou Xiaoye,Anguo-1,Anguo-2 and Anguo-3 increased firstly and then decreased.Compared with the control,Bozhou Xiaoye increased the most;the antioxidant enzymes activities of five varieties of Isatidis Radix seedlings increased first and then decreased.Compared with the control,Bozhou Xiaoye increased the most,followed by Anguo-1,Bozhou Daye increased the least;when salt concentration was 40 mmol/L,MDA content of Bozhou Xiaoye did not change significantly compared with the control,and the other four varieties MDA content increased significantly,MDA content of five varieties increased significantly under high salt concentration;proline content of Isatidis Radix of five varieties continued to increase.Compared with the control,Bozhou Xiaoye increased the most;soluble sugar content of Isatidis Radix of five varieties increased in varying degrees.Compared with the control,Bozhou Xiaoye increased the most and Bozhou Daye increased the least,under high salt concentration,respectively.The increase range of all varieties decreased.Through the comparison and analysis of the physiological indexes of the five varieties of Isatidis Radix seedlings,it was found that Bozhou Xiaoye had the strongest salt-alkali resistance,followed by anguo-1,which was consistent with the salt and alkaline tolerance of seeds during germination stage.
