In order to solve the problem of seed dormancy restriction in the cultivation and production of red peony root in northeast China,theoretical basis and reference were provided for the efficient cultivation and production of artificial red peony root.In this paper,the seed germination stage of red peony root was studied,and the changes of seed germination under different treatment conditions were explored,so as to find a reasonable regulation scheme for seed germination of red peony root.In this study,the experimental environment of artificial cultivation of red peony root seeds in laboratory was simulated.Seeds of red peony root were treated with conventional stratification,variable temperature stratification and different concentrations of GA3 and CTK hormones.The rooting and germination of red peony root seeds under different treatment conditions,as well as the changes in nutrient content and enzyme activity during germination were measured.To explore the method to break the hypocotyl dormancy of red peony seed.The optimal treatment conditions were summarized to break the dormancy of red peony seeds and improve the germination rate of red peony seeds.Through the analysis and comparison of the rooting initiation date,rooting rate,rooting index,mildew and rot rate,taproot length and diameter of hypocotyl of red peony root seeds treated by conventional stratification and variable temperature treatment,it can be seen that the growth of red peony root seeds treated by conventional stratification is superior to that of variable temperature stratification.Through stratification treatment of red peony root seeds,it was found that under conventional stratification treatment of 60 d,seed germination time was short,germination rate was high,nutrient content changes were more significant,and POD and CAT activities were effectively increased.Secondly,variable temperature stratification treatment with low temperature of-20?for 40 d and stratification treatment with 5?for 20 d can also promote the growth of red peony root seeds.Through the comparison and analysis of different concentrations of GA3 and CTK hormone in seeds of red peony root,it was found that at the concentration of 20 ppm,it was more helpful to break the dornidity of seeds of red peony root,with the occurrence rate as high as 67.7%and71.4%,respectively.The increase rate of seeds of red peony root under the concentration of CTK of 20 ppm was better than that under other concentrations.All the above test results,in the northeast of 15?to 25?greenhouse environment,radix paeoniae rubra seeds to 5?conventional laminated processing 60 d,as well as the concentration of 20 ppm CTK solution processing,also can make the seeds of radix paeoniae rubra shorten the cultivation time of 3 months,early stage of the thief,promoting the transformation of nutrients,helps to break the hypocotyl herbaceous peony seed dormancy,improve biological protective enzyme activity,enhance the rooting rate and radix paeoniae rubra seeds germination rate.
