In the research of animal and plant inspection,forensic evidence identification,species phylogeny,disease diagnosis,etc.,it is not known that the species identification of biological samples is an important identification link.Traditional species identification methods rely on morphological methods and anatomy.Methods and chemical analysis,but due to environmental factors and differences in different stages of biological development,these methods show limitations.In order to overcome these shortcomings,many molecular identification methods based on genotyping have been published.In this paper,DNA barcodes of scorpions were constructed by internationally used plant DNA barcode primers(ITS2,matK,rbcL and psbA-trnH),and HPLC fingerprints were established to identify scorpions from various aspects.Provide basis for breeding of sub-medicinal varieties,carry out standardization,and obtain the following results:1.According to the field observation,in response to the differences in plant morphology,leaf morphology,fruit shape,color and other traits,the fruit and leaves of the scorpion were collected in the main scorpion producing areas of Hunan,Jiangxi,Chongqing,etc.,and the morphological characteristics of the fruit were carried out.Comparison.2.The DNA of the leaves of the scorpion was extracted by the kit method.The PCR was amplified by four primers,ITS2,matK,rbcL and psbA-trnH.The amplified products were detected.The analysis showed that the rbcL amplification product can be used as a characterization.The primary barcode of scorpion,matK,ITS2,psbA-trnH amplification products are used as second,third and fourth bar codes respectively,and the scorpion population is subdivided step by step,reflecting the genetic diversity of scorpion population.3.Dry and smash the hazelnut fruit and pass through a 40 mesh sieve.The methanol extract was tested by HPLC fingerprint.The results showed that the relative retention time(t)of the common peaks of the fingerprints of different gardenia medicinal materials was basically the same,reflecting The main components contained in the medicinal materials are similar,but the relative peak area(A)has a certain difference,reflecting the difference in the content of the main components of different medlars.The similarity of 18 HPLC fingerprints of 20 kinds of Gardenia medicinal materials was above 0.9,and the similarity of S15 and S20 was lower,and the similarity was only 0.845 and 0.867.The results of cluster analysis are consistent with the results of similarity analysis.4.The DNA barcode information of Gardenia scorpion reflects the commonality and diversity of the genetics of Gardenia sinensis.The HPLC fingerprint information of Gardenia sinensis reflects the commonality and difference of the chemical constituents of Gardenia sinensis.Genetic stability;the authenticity of the medicinal material is based on the optimization of the chemical composition of the genetically stable germplasm material under different habitat conditions to achieve optimal drug efficacy;therefore,based on DNA barcode and HPLC fingerprint information The selection of "Dragon-code" combined with the control of the quality of the medicinal materials has a good guiding significance.
