| Objective: To establish a bone metastasis model of breast cancer in nude mice,observe and study the effect of Bushen Huoxue decoction on breast cancer bone metastasis model and Jagged1/Notch and TGF-? pathway proteins in nude mice.Methods: 56 female 4-5 week-old female nude mice were stratified according to physical quality and divided into model group,zolead phosphate group,blank group,high concentration of Bushen Huoxue Huoxue,Bushen Huoxue Decoction concentration group,Bushen Huoxue Decoction.Low-dose group,kidney-tonifying group,and blood-activating group 8 groups,7 in each group.The breast cancer bone metastasis model was constructed in each group.The blank group was not treated.The construction steps were as follows:(0.1 ml,1×107/ml)human breast cancer MDA-MB-231 cell suspension was infused into the left ventricle of nude mice.A bone metastasis model for breast cancer was constructed.After the model was successfully constructed,the physical changes in nude mice were observed and recorded.On the second day after the successful replication of the model,each group was given drug administration for 6 weeks,and the blank group was not treated.The specific drug intervention was as follows: The mice in the low,middle,and high concentrations of kidney and blood were given raw drug concentrations of 10 g in order./kg,20g/kg,40g/kg daily gavage;Huoxue group(bush kidney activating blood soup deconstructed party)given raw drug concentration 0.4g/kg daily gavage;Bushen group(bush kidney activating blood soup disassembled party)given crude drug concentration 2 g/kg daily gavage;zoledronic acid group was given intraperitoneal injection of 100 ?g/kg once a week;model group was given 0.2 ml NS daily gavage.On the 40 th day after model establishment,nude mice were subjected to radionuclide bone ECT scanning to record the bone metastases of nude mice in each group.On the 42 nd day,the nude mice were sacrificed by abdominal aorta and the nude mice were killed by cervical dislocation.All groups of diseased bone tissue were removed on a sterile working table to confirm pathological diagnosis of bone metastases.HE staining was used to observe the degree of bone destruction in each group.Immunohistochemical method was used to detect the expression of Jagged1 and TGF-? protein in each bone tissue.Results: 1.After injecting human breast cancer MAD-MB-231 cells into the left ventricle of nude mice,a bone metastasis model of breast cancer can be successfully constructed.2.After the model was constructed,the bone metastasis rate in the model group was 100%,and the rest of the bone metastases were inhibited to varying degrees.3.The physical quality of the nude mice in the model group was significantly lower than that in the treatment group,and the difference between the groups was statistically significant(P<0.05).4.The expression of Jagged1 in the high concentration group of Bushen Huoxue was lower than that of the zoledronic acid group,and the difference between the two groups was statistically significant(P<0.05).The expression of TGF-? in the high concentration group of Bushen Huoxue was significantly lower than the other groups(P<0.05).There was a statistically significant difference between groups.The expression of Jagged1 and TGF-? in the model group was significantly higher than that in the treatment group.The difference between the two groups was statistically significant(P<0.05).Conclusions: 1.After injecting human breast cancer MAD-MB-231 cells into the left ventricle of nude mice,a bone metastasis model of breast cancer can be successfully con structed.2.Bushen Huoxue Decoction can effectively reduce and inhibit the occurrence of bone metastasis.3.Bushen Huoxue Decoction can enhance the body weight of nude mice and improve the nude mice's constitution.4.Bushen Huoxue Decoction may inhibit the expression of Jagged1 and TGF-? and interfere with the Jagged1/Notch and TGF-? pathways,and inhibit the bone metastasis of breast cancer. |
