Objective:To study the effect of Huxinkang on cox-2 m PGES-2 in Apo E-r-atherosc-lerosis mice.Methods:Eight C57 BL / 6 mice were used as wild control group.38 Apo E-r-R mice were fed with normal diet.They were randomly divided into 8 Apo E-r-control group(n = 8)and as model group(n = 30).The rest were fed with high-fat diet(21% fat and0.15% cholesterol).After 13 weeks and 13 weeks of modeling,two thoracic aorta were randomly selected from the model group to observe their thoracic aorta.Four mice died in the course of modeling.According to the random number method,they were divided into the following groups: model group,Huxinkang group,Atto group.After success,the oral dose of Huxinkang group was 43.2 g / kg ·d,and that of Atto vastatin control group was 3 mg / kg ·d.The gavage volume was 0.5 ml / d.After all the drugs were mashed and dissolved in distilled water,the suspension was prepared and the volume of stomach was 0.5 ml / d / d.The histopathological changes of aorta were observed under high power microscope after 4 weeks and 17 weeks of HE staining.The expression of cox-2m PGES-1 protein was detected by IHC method.Results:1.The pathological morphology of aorta was observed with naked eyes: the aortic intima of the two control groups were relatively smooth,the subintimal structure was relatively complete,the intima was not thickened,plaque was not formed,and no obvious lumen stenosis was observed.The aortic intimal wall was irregular in the model group.Its association is not strong,obvious thickening,large degree of stenosis in the lumen,and wide area,plaque protruding into the blood vessels,forming a typical atherosclerotic plaque,can be seen thin fibrous cap,some fibrous cap edge is diversity,irregular.Discontinuous,large lipid core,accumulation of large amounts of cholesterol crystals and adipocytes.Although there are a few plaques in the Huxinkang group and Atto vastatin group The content of foam cells was less than that of model group.Conclusion:1.Huxin Kang could significantly improve the histopathological changes of aorta and reduce the plaque area ratio in as mice.2.Huxinkang can decrease the expression of cox-2m PGES-1,delay the development of atherosclerotic plaque,reduce the area of atherosclerotic plaque,increase the patency of artery,and stabilize the atherosclerotic plaque.
