ObsjectiveThe purpose of this study is to investigate the effect and mechanism of effective composition of Salvia miltiorrhiza Bunge on the inflammatory response of cerebral ischemia reperfusion injury based on TCM prescription and NLRP3-caspase-1 pathway.MethodThis expriment uses MCAO method to make rat cerebral ischemia-reperfusion injury model.SD rats were randomly divided into 6 groups,10 in each group,sham operation group,model group,nimodipine group(0.5mg/kg),Danshensu(2 mg/kg),hydroxysalvanin A group(2 mg /kg),Danshensu(1mg/kg)compatible with hydroxysaccharin A(1mg/kg)(1:1).Zealon,s method was used to perform neurological scores in rats;TTC staining was used to observe and calculate cerebral infarct area;serum SOD and NO levels were measured using kits;pathological changes of brain were observed by HE staining;brain tissue NLRP3 was detected by immunohistochemistry.And caspase-1 expression;Elisa assay serum 1L-18 and IL-1?.ResultStudies on the compatibility of Danshensu Hydroxysingophyllin A(1:1)with cerebral ischemia-reperfusion injury in rats showed that:1.The neurological scores of the rats in the treatment group and the model group were improved,and the inflammatory infiltration was alleviated by HE staining.2.From the expression of NLRP3 and caspase-1 in rat brain and the expression of IL-1?and IL-18 in serum,the content of NLRP3 in model group and sham group was increased(P<0.05),and the content of caspase-1 was increased.(P<0.05),the content of IL-1?increased(P<0.05),and the content of IL-18 increased(P<0.05).There was a significant difference;the treatment group and the model group could reduce the content of NLRP3 in brain tissue.(P<0.05)or(P<0.01),decreased caspase-1 content(P<0.05),decreased IL-1?,IL-18 content(P<0.01);significantly reduced brain tissue;Danhong compatibility group The expression of NLRP3 in the Danshensu group and hydroxycarmelene A group decreased(P<0.01),the expression of caspase-1 decreased(P<0.05),and the expression of IL-1? and IL-18 decreased(P<0.01).3.From the results of serum SOD and NO expression,Danhong compatibility group and Danshensu and hydroxycarmelene group A increased the serum SOD expression(P<0.05)and reduced the NO expression in serum(P<0.05);Salvia miltiorrhiza Compared with nimodipine group,serum SOD level was higher in prime group(P<0.05),and serum NOlevel was lower in hydroxyseloerylanin A group and nimodipine group(P<0.05).ConclusionThe experimental results show that the combination of Danshensu and hydroxysafflowerA(1:1)can protect against cerebral ischemia-reperfusion and reduce the inflammatory response in cerebral ischemia-reperfusion injury.The mechanism may be related to the down-regulation of NLRP3-caspase-1 signaling.The pathway is associated with the expression of NLRP3,caspase-1,1L-18,and IL-1?.
