Objective:To discusses the effect of electroacupuncture Back-shu and Front-mu on the gastrointestinal transmission function,colon tissue morphology and colon ICC,SMC in functional constipation in mice,aims to explore the mechanism of acupuncture large intestine Back-shu and Front-mu acupoints on the disease,and to provide the scientific basis for the clinical selection and application of acupoint for acupuncture treatment of this disease.Method:1.Experiment one: 80 Kunming mice were selected and randomly divided into 5groups: blank control group,model group,blank acupuncture group,acupuncture group and drug group,with each group having a total of 16 mice.Using gavaging compound diphenoxylate for mice to make constipation model,chooseing bilateral Tianshu and dachangshu to alternate electroacupuncture,30 min every time,1 times a day,5 times for a course of treatment,2 days of rest,a total of 2 courses of treatment.After first courses of treatment and the end of the second course of treatment,then fasting 12 h,activated carbon by gavage at a dose of 0.1ml/10 g,after 30 minutes mice were killed,clipping the bowel,measuring the rate of small intestine,while clipping the mice of distal colon,detection the colonic smooth muscle tension changes.2.Experiment two: 80 Kunming mice were randomly divided into 5 groups:blank control group,model group,blank acupuncture group,acupuncture group and drug group,with each group of 16 rats in each group.The methods of model making and electro acupuncture were all the same as experiment one.Animals in each group were sacrificed at the end of the experiment,and then the distal colon was removed.A part of the colonic tissue was used to observe the colon morphology of MOFT BA600 Mot automatic digital high-power scanning microscope.The ICC and SMC cells were stained with c-kit and ?-SMA,and then stained with TCS-SP2 laserconfocal microscopy.The images were collected and the images were collected,then were reconstructed by Image-ProPlus 6.0 image analysis software and the fluorescence values were quantitatively analyzed.Results:1.gastrointestinal transmission function test(1)small intestinal propulsive rateCompared with the blank control group,the small intestinal propulsion rate of the model group was significantly reduced,the difference was statistically significant(P<0.01),suggesting that the small intestine propulsion speed of functional constipation mice slowed down.Compared with the model group,acupuncture group and drug group of small intestinal propulsion rate was significantly increased,the difference was statistically significant(P<0.05,P<0.01),suggesting that acupuncture and drugs can significantly speed up the functional constipation mice intestinal propulsive rate.Compared with blank acupuncture group,the small intestinal propulsive rate of the blank control group was not significant(P > 0.05),suggesting that the effect of acupuncture on the intestinal propulsive rate was not obvious.There was no significant change in intestinal propulsive rate in each group(P > 0.05)between the two groups.(2)the percentage of average tension of colonic smooth muscleCompared with the blank control group,model group mice of colonic smooth muscle tension amplitude decreased,the difference was not statistically significant(P > 0.05);compared with the model group,acupuncture can enhance the tension of colonic smooth muscle changes in constipation mice(P < 0.05);the blank control group was compared with the blank acupuncture group,there was no significant difference between the two group,the tension of colonic smooth muscle changes percentage(P > 0.05).There was no significant change in the smooth muscle tension between the two groups(P> 0.05).2.colon tissue morphology and ICC,SMC cells confocal laser scanning(1)observation of colonic morphologyBlank control group normal colonic smooth muscle thickness,smooth muscle cells arranged in neat,morphologically normal colonic mucosa,submucosal gland is rich and neat,no edema and hyperemia,muscle layer in the myenteric plexus of a larger number of nerve cells arranged in neat;model group: colon smooth muscle layer thickness,smooth muscle cells arranged in disorder cells,vacuolar degeneration,colonic mucosal integrity is poor,acinar gland atrophy,reduced,sparse,irregular arrangement,no edema and hyperemia,myenteric plexus to reduce the number of nerve cells arranged in disorder,see vacuolar changes;blank acupuncture group,acupuncture group and drug group colon morphology and blank control there is no significant difference between groups.Compared with the two courses,the histomorphological changes of colon tissue in each group were not obvious.(2)ICC and SMC cells were observed by confocal laser scanningCompared with the blank control group,the fluorescence intensity expression of c-kit and alpha-SMA were decreased significantly(P < 0.01,P < 0.01)in model group,the fluorescence intensity expression of c-kit were decreased significantly(P <0.01)and the expression of alpha-SMA fluorescence intensity were decreased(P <0.05);compared with the model group in the blank acupuncture group,in the acupuncture group and the medicine group,the expression of c-kit fluorescence intensity were enhanced(P < 0.05,P < 0.05),acupuncture group alpha-SMA the expression of fluorescence intensity were enhanced(P < 0.05),drug group alpha-SMA expression of fluorescence intensity increased,the difference was not statistically significant(P > 0.05).Compared with the two groups,there was no significant change in the expression of c-kit and alpha-SMA in colon of each group(P > 0.05,P > 0.05).Conclusion:1.The effect of acupuncture on the gastrointestinal transmission function,colon tissue morphology and colon ICC,SMC cells was not obvious.2.Acupuncture and moxibustion can improve the gastrointestinal transit function of the functional constipation mice,colonic tissue morphology,increase the immuneactivity of ICC and SMC in mice,promote the recovery of ICC and SMC,and improve the gastrointestinal motility,Treatment of functional constipation.3.The curative effect of acupuncture on the treatment of functional constipation in the treatment of functional constipation is not proportional versus the course of acupuncture treatment.
