Objective:Through SCF/c-kit signaling pathway,the effect of Cistanche deserticola on interstitial cells of Cajal in rats with functional constipation was studied.Methods:Twenty-four 8-month-old SPF SD male rats were randomly divided into four groups,six rats in each group.During the film period,rats in the model,treatment,and blocking groups were intragastrically administered with loperamide(0.063mg/100g,10ml/kg),and the first dose was doubled and administered once a day for 20 consecutive days:the normal group was intragastrically administered with the same amount of saline.At the same time of modeling,drug differentiation treatment was performed for each group:the normal group was given normal saline by gavage once a day for 20 consecutive days;the model group was given normal saline by gavage once a day for 20 consecutive days;the treatment group was given Cistanche deserticola(0.158g/100g,10ml/kg)by gavage once a day for 20 consecutive days;the blocking group was given Cistanche deserticola(0.158g/100g,10ml/kg)by gavage once a day for 20 consecutive days,and at the same time,intraperitoneal injection of STI571(25 mg/ml,1 ml/kg)for 20 consecutive days.The general condition,stool particle number,stool weight and stool quality of the four groups were observed.The expression of Cajal interstitial cells,c-kit protein and c-kit gene was detected by HE staining,immunohistochemistry and RT-PCR.Results:Compared with the normal group,the body weight,stool particle number and stool weight of the rats in the model occlusion group were significantly reduced(P<0.05),and the stool water was reduced.The body weight,stool particle number,stool weight and stool water of the rats in the treatment group were slightly reduced compared with the normal group(P<0.0 5),but higher than those in the model occlusion group.There was no significant difference between the model occlusion groups(P>0.05).Compared with the colon tissue of rats in the model group,the inflammatory changes of rats in the treatment group were reduced,the c-kit positive area was increased(P<0.05),and the c-kit mRNA expression was increased,while there was no significant difference in the blocking,with no statistical significance.Conclusion:Cistanche deserticola can improve the symptoms of functional constipation rats and increase the activity of interstitial cells of Cajal,and the possible mechanism is that Cistanche deserticola affects interstitial cells of Cajal through SCF/c-kit signaling pathway and improves colonic sensitivity.
