| Objective1.By establishing a mouse model of cervical cancer-bearing tumor,to explore the anti-tumor effect of traditional Chinese medicine Qingdu suppository;2.To investigate the effect of Chinese medicine Qingdu suppository on the levels of serum INF-y,TGF-?1 and IL-10 in cervical cancer-bearing mouse,and to analyze the anti-tumor mechanism ofQingdu suppository;3.Through the use of traditional Chinese medicine inheritance platform,the Chinese medicine treatment of HR-HPV infection in the cervix is deeply explored,which provides a reference for clinical treatment.MethodExperimental Study1.Establish mousemodels of cervical cancer-bearing tumor.After successful modeling,they were randomly divided into model control group,low-dose Chinese medicine group and high-dose Chinese medicine group,Qingdu suppository was treated by continuous gavage for 28 days.On the 3rd and 7th day of each week,the body weight and tumor volume of each group of mice were measured and recorded.On the 28th day,the tumor tissue was removed,the tumor weight was measured,and the tumor inhibition rate of the Chinese medicine group was calculated.2.0n the 28th day,the tumor tissues of each group were taken for paraffin section and HE staining,and to observe the effect of Qingdu suppository on tumor cell proliferation and apoptosis.3.On the 28th day,peripheral blood of each group of mice was taken,and to detectthe levels of INF-?,TGF-?1 and IL-10 in peripheral serum ELISA.literature ResearchRetrieved from January 1,2008 to December 31,2008,published in the China Knowledge Network,Wanfang Chinese Journal Database on Chinese medicine for the treatment of cervical high-risk human papillomavirus(HR-HPV)infection,using Chinese medicine Inherit the auxiliary platform for data analysis and mining,and initially obtain the rules,core combinations and alternative new prescriptions for the treatment of cervical HR-HPV infection.Result1.Compared with the cervical cancer model control group,the tumor volume of the low-dose drug group and the high-dose drug group was significantly reduced,the tumor weight was also significantly decreased,and the high-dose group was down-regulated more significantly(compared with the low-dose group,p<0.01).There was no significant difference in body weight during the entire experimental period in the low-dose drug group.From the 7th day,the mice in the high-dose drug group showed a significant increase in body weight(p<0.05)compared with the cervical cancer model control group and the low-dose drug group.2.Compared with the cervical cancer model control group,the serum INF-? levels in the peripheral blood of the low-dose and high-dose groups were significantly increased(P<0.05).The serum INF-? content in the high-dose drug group was significantly higher than that in the low-dose drug group,and the difference was statistically significant(P<0.01).Compared with the cervical cancer model control group,the levels of TGF-?1 and IL-10 in peripheral blood of two drug-group mice were both significantly decreased(P<0.01),and the levels of TGF-?1 and IL-10 in the high-dose group were significantly lower than those in the low-dose group.The difference was statistically significant(P<0.01).3.HE staining results:The tumor cells in the tumor tissues of the cervical cancer control group were arranged in a disordered manner,the cell size was uneven,and the nuclear granules were less and mitosis was observed.After low-dose treatment,the tumor tissue of the mice was unclear and regional necrosis,interstitial edema and inflammatory cell infiltration;after high-dose treatment,the tumor tissue of the mouse was unclear and large-area regional necrosis was observed.Edema and nuclear concentration,different depths,a large number of inflammatory cells infiltration.4.Analysis of the rule of medication for oral administration of the Cer vical HR-HPV infection:a total of 59 prescriptions in accordance with the inclusion criteria,115 Chinese medicines,the order of the top ten used are:White Atractylodes rhizome,Semen coicis,Astragalus,Cortex phellodendri,Oldenlandia,Licorice,Codonopsis,Rhizoma smilacis glabrae,Atractylodis,Tuckahoe;the frequency of drug use decreased in the cold,warm,flat,and cool,of which cold drugs dominate.The medium-frequency frequency of the five flavors is Gan,bitter and Xin.In the statistics of drug return,the top three frequencies were spleen,liver and stomach,and the core combination was 12,and 6 new prescriptions were introduced.5.Analysis of the regularity of Chinese medicine for external treatment of cervix HR-HPV infection:A total of 46 prescriptions,85 Chinese medicines,which were in accordance with the inclusion criteria,were used in 10 or more times,followed by Cortex phellodendri,Bomeol,Sophora flavescens,Lithospermum,and Rhizoma Curcumae.The frequency of drug use decreased in the order of cold,warm,flat and cool,of which cold drugs dominated.The medium-frequency frequency of the five flavors is Gan,bitter and Xin.The top three frequencies of drug use are spleen,liver and stomach.A total of 10 core combinations were obtained,and 5 new prescriptions were introduced.Conclusion1.Chinese medicine Qingdu suppository can significantly reduce the tumor volume and weight of cervical cancer-bearing mice,and Qingdu suppository can inhibit tumor growth.2.Qingdu suppository can inhibit the proliferation of tumor cells in tumor tissues of tumor-bearing mice and promote the apoptosis of tumor cells,thus playing a therapeutic role,and the higher the concentration,the more significant the curative effect.3.Qingdu suppository can up-regulate the level of INF-?,reduce the content of TGF-?1 and IL-10,and has certain anti-viral effect,which may reverse the immunosuppression in the micro-environment,and further experimental verification is needed.4.Oral Chinese medicine for treating HR-HPV infection of the cervix is mainly based on Fuzheng Buxu combined with clearing away heat and detoxifying,clearing away heat and dampness drugs,reflecting the combination of Fuzheng and Qixie;the topical drugs for treating HR-HPV infection in the cervix are mainly heat-clearing.Detoxification and cooling blood drugs are the main drugs,and the drugs with higher frequency are Huangbai,borneol,Sophora flavescens,comfrey,and phlegm,which provide reference for clinical drug use. |
