Objective:To investigate the effects of Nao Shuming decoction on nerve regeneration and GAP-43 and BDNF expression in rats with focal cerebral ischemia.Methods: 300 SD rats were used to feed the general laboratory of the First Affiliated Hospital of Hunan University of Traditional Chinese Medicine.The rat model of focal cerebral ischemia was replicated by middle cerebral artery occlusion(MCAO).The Longa 5 method was performed 2 hours after operation.Neurobehavioral scores,1-3points were included in the study.After successful modeling,the model group and the Nao Shuming decoction high,medium and low dose groups were randomly divided into groups of 54 each.On the 3rd,7th,and 14 th postoperative day,18 rats died in each group.The Ayelet Levy 14 method was used to score the nerve function.The TTC staining was used to measure the area of cerebral infarction.Morphological and immunohistochemical methods were used to detect the expression of GAP-43 and BDNF on the ischemic side of rats.Detection of GAP-43 and BDNF gene expression by qPCR.Results:1.Compared with the model group,The neurological function scores of the high,middle and low doses decreased at 7d and 14d(P<0.05),The effect was most pronounced in the middle dose group.2.Compared with the model group,3d,7d,14 d cerebral high,medium and low dose groups can reduce the infarct size(P<0.05),compared with the 3-day cerebral sputum medium dose,7d,14 d cerebral palsy The dose significantly reduced the infarct size(P<0.01).3.Compared with the model group,the expression levels of GAP-43 in the high,middle and low groups of Nao Shuming decoction increased at 3d,7d and 14 d,the difference was statistically significant(P<0.05);The positive expression of GAP-43 was significantly increased in the middle group after 14 days of modeling(P<0.01).4.Compared with the model group,the expression of BDNF in the brain tissue of rats in each dose group was increased at 3d,7d and 14 d after treatment with Naoshuming Decoction(P<0.05),and the expression of BDNF was significantly increased in the middle dose group of Nasoshuming at 14 d.<0.01).5.Compared with the normal group,the expression of BDNF in the brain of model rats was increased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of GAP-43 in the brain tissue of each group was increased after 3 days,7 days and 14 days after treatment with different doses of Nao Shuming.The difference was statistically significant(P<0.05).The expression of GAP-43 was significantly increased in the middle dose(P<0.01).6.Compared with the model group,the expression of BDNF gene in the brain tissue of each group was increased at 3d,7d and 14 d after treatment with Naoshuming Decoction,the difference was statistically significant(P<0.05),and the expression of BDNF gene was expressed at 14 days.Significantly enhanced with significant differences(P < 0.01).Conclusions:1.Nao Shuming decoction can reduce the neurological function score of MCAO rats,reduce the area of cerebral infarction to promote recovery.2.Nao Shuming decoction may regulate the expression of BDNF and GAP-43,up-regulate the expression of GAP-43 and BDNF mRNA to protect brain tissue in cerebral ischemia.
