Objective:To study the Xijiao Dihuang Decoction's brain protection to the focal cerebral ischemia reperfusion rats and effects on the angiogenesis and nerve regeneration associated proteins expression.To provide experiment basis and pharmacological basis for the treatment of cerebral infarctionMethods:Healthy male SPF SD rats were assigned randomly to 5 group:Sham group,Model group,Nim group,XJDH-H group and XJDH-L group.The animals were given medicine every day in 1 weeks before modeling.XJDH-H group and XJDH-L group were respectively administered with Xijiao Dihuang Decoction at dose of 22.5g/kg·d?11.25g/kg·d.Nim group was administered with Nimodipine at dose of 1mg/kg-d.Sham group and model group were given equal volume of normal saline.The operation model were established 2 hours later after the last administration.The focal cerebral ischemia reperfusion rats models were established by middle cerebral artery occlusion.The rats of ischemia reperfusion were realized after 2 hours ischemia.24 hours later after being awake,the neurological function scores of rats were examined and the nerve cells structure change,infarction volume were observed by HE and TTC staining.The expression of VEGF,bFGF,CD34,BDNF,GDNF in ischemia brain tissue were examined by Western blot analysis.Results:1.The Xijiao Dihuang Decoction's effects on the neurobehavioral of focal cerebral ischemia reperfusion rats.The neurobehavioral of Sham group had no change,which was scored 0.Compared with Sham group,the neurological function score of Model group had significantly difference(p=0.000).Compared with Model group,XJDH-H group can effectively decrease the neurological function scores at 2d,3d,4d(p<0.01).XJDH-L group can effectively decrease the neurological function scores at 2d(p<0.01).Nim group can effectively decrease the neurological function scores at 2d,3d(p<0.01,p<0.05).2.The Xijiao Dihuang Decoction's effects on the brain histopathology of focal cerebral ischemia reperfusion rats.The results of histomorphology showed:the nerve cells structure of the sham group was regular and closely connected.The model group had serious change in nerve cells structure:disorder cell structure,different size,deeply stained nuclei and condensation,unclear nucleolus and membrane,widened gap around nerve cells.The neuronal ischemic changes of the treatment groups were ameliorated to a certain extent.XJDH-H group could significantly ameliorate the above-mentioned changes.3.The Xijiao Dihuang Decoction's effects on the infarction volume of focal cerebral 'ischemia reperfusion rats.Sham group had no infarction.Compared with Sham group,obvious cerebral infarction could be seen in the model group.Compared with Model group,the cerebral infarction volume of XJDH-H group and Nim group was obviously reduced(p<0.01,P<0.05)and XJDH-H group was more effective than the Nim group(P<0.05).But cerebral infarction volume of XJDH-L group was not obviously reduced(P>0.05).4.The Xijiao Dihuang Decoction's effects on the angiogenesis regeneration associated proteins expression of focal cerebral ischemia reperfusion rats.In the Sham group,the expressions of VEGF,bFGF and CD34 were low.Compared with Sham group,the expressions of VEGF and bFGF was increased in model group(p<0.05),but the expressions of CD34 had no significant difference(p>0.05).Compared with Model group,the expressions of VEGF,bFGF and CD34 was obviously increased in XJDH-H group and XJDH-L group(p<0.05,p<0.01),and the effect was in a dose-dependent manner.Compared with Model group,the expressions of VEGF,bFGF and CD34 could increase observably in Nim group(p<0.01).5.The Xijiao Dihuang Decoction's effects on the nerve regeneration associated proteins expression of focal cerebral ischemia reperfusion rats.Compared with Sham group,the expressions of BDNF and GDNF was not obviously increased in model group(p>0.05).Compared with Model group,the expressions of BDNF and GDNF was obviously increased in XJDH-H group and Nim group(p<0.01).Compared with Model group,the expressions of BDNF was increased in XJDH-L group(p<0.05),but the expressions of GDNF had no significant difference(p>0.05).Conclusion:Xijiao Dihuang Decoction can improve the brain histomorphology,reduce the area of cerebral infarction and has a protective effect on focal cerebral ischemia reperfusion rats,Which may be related to upregulate the expressions of VEGF,bFGF,CD34,BDNF,GDNF in ischemia brain tissue.
