Objective:To observe the gastrointestinal tract function of rats with slow transit constipation(STC)and the morphological structure of Enteric nervous system-Cajal interstitial cells-Smooth muscle cells(ENS-ICC-SMC)in rats with electroacupuncture Shu and Mu acupoints in large intestine.The impact of this is to provide a scientific basis for the treatment of STC by electroacupuncture Shu and Mu acupoints in large intestine.Methods: The 64 SPF rats were randomly divided into 16 cases in blank group and16 cases in saline group and 32 cases in model group.The model was made by compound diphenoxylate gavage method to replicate the STC model.The saline group was administered with the same dose of normal saline.The blank group was routinely fed and not treated.After successful modeling,the model was randomly divided into16 model groups and 16 electroacupuncture groups.The electroacupuncture group was treated with bilateral Shu and Mu acupoints in large intestine at the same time.Each electroacupuncture treatment was performed for 15 minutes,once a day for 14 days.The other groups were fixed in the same method for 14 days.Observe the general condition of the rats in each group and the changes of feces.Changes of gastrointestinal transit function in rats in each group were detected by black semi-solid paste propulsion method.Observation of distal colon tissue cholinergic nerve and ICC by immunofluorescence laser scanning confocal microscopy;The ultrastructure of colon tissue ENS-ICC-SMC was observed by transmission electron microscopy.Result:1.Comparison of general conditions in rats: There was no significant difference in the general condition between the blank group and the saline group,all of them showed fur luster,normal diet and good mental state.The rats in the model group were fluffy,dull,with less food and less susceptibility.It is struggling and struggling,and the mental state is not good.The rats in the electroacupuncture group have thesame pre-treatment condition as the model group.After treatment,the hair is brighter than before,the eating condition and mental state are improved,and the struggling force is less than before.2.Comparison of 24-hour defecation weight and fecal trait score in rats:Before treatment,the model group and the electroacupuncture group were statistically significant compared with the blank group and the saline group(P<0.05),suggesting that STC rats have reduced bowel movements and symptoms of fecal dryness;The defecation and fecal trait scores of the electroacupuncture group were similar to those of the blank group and the saline group(P>0.05),and were significantly higher than before treatment(P<0.05),suggesting that electroacupuncture Shu and Mu acupoints in large intestine can effectively improve the symptoms of less defecation and dry stool in STC rats.The model group was improved after treatment(P<0.05),but the difference was statistically significant compared with the other three groups(P<0.05),suggesting that the STC model rats have a certain degree of self-healing after a period of time,but Still worse than normal.3.Comparison of gastric emptying rate and small intestine propulsion rate:Compared with the blank group:the difference between the saline group and the electroacupuncture group was not statistically significant(P>0.05);the difference between the model group was statistically significant(P<0.05).Compared with the saline group:the difference between the model group was statistically significant(P<0.05);the difference between the electroacupuncture group was not statistically significant(P>0.05).Compared with the model group: the electroacupuncture group was significantly increased,and the difference was statistically significant(P<0.05).It is suggested that the gastric emptying rate and the small intestine propulsion rate of STC rats are decreased,and electroacupuncture Shu and Mu acupoints in large intestine can significantly increase the gastric emptying rate and intestinal propulsion rate of STC rats,thus improving the gastrointestinal transit function.4.Immunofluorescence laser scanning confocal microscopy observation the cholinergic nerve and ICC:Blank group: ChAT and c-kit have normal fluorescence intensity,cholinergic nerve-ICC has normal and uniform distribution,and is closely related;Saline group: ChAT and c-kit fluorescence intensity are compared with blank group.There was no statistical significance(P>0.05).The morphological structure ofcholinergic nerve-ICC was basically normal,and the distribution was relatively uniform and closely related.Model group : The fluorescence intensity of ChAT and c-kit in the model group was significantly lower than that in the blank group and the saline group(P<0.05).The morphological structure of cholinergic nerve-ICC was ambiguous and unevenly distributed,and the relationship was not tight or discontinuous.Electroacupuncture group: The fluorescence intensity of ChAT and c-kit was not significantly different from that of the blank group and the saline group(P>0.05),but the difference was statistically significant compared with the model group(P<0.05).The cholinergic nerve-ICC has improved morphology,clear structure and close connection.It indicated that the expression of ChAT and c-kit in STC rats was significantly decreased,and the morphological structure of cholinergic nerve-ICC was damaged.Electroacupuncture at the site of large intestine decoction could alleviate the damage of the choline-structure of distal colonic cholinergic nerve-ICC in STC rats.It is restored to a more complete.5.ENS-ICC-SMC network is observed by transmission electron microscopy:the ultrastructure of the blank group ENS-ICC-SMC network is normal;the ultrastructure of the saline group network is basically normal;the ultrastructure of the model group network is destroyed;the ultrastructure of the electroacupuncture group network Restored to normal.Conclusion:1.Electroacupuncture Shu and Mu acupoints in large intestine can improve the general condition,fecal condition and gastrointestinal transit function of STC rats.2.Electroacupuncture Shu and Mu acupoints in large intestine can improve the ultrastructure of the ENS-ICC-SMC network in STC rats,thereby improving gastrointestinal motility.
