Objective:To observe the effect of pathological changes and the content of VIP, SP and 5-HT in the lungs and colon of STC rats, and the effect of treatment on STC of rats with the Treating lung method, Treating large intestine method and Treating lung and large intestine simultaneously.To explore the mechanism of treatment on STC with the Treating lung and large intestine simultaneously which is based on Treating large intestine disease from lung. To provide the animal experience basis for the developing new drug which is treating the STC based on lung theory. And in order to further explore the theory which is "The lung and the large intestine being interior-exteriorly related". Methods:50 SD rats were randomly divided into 5 groups:Blank control group, Model control group,Treating lung group, Treating large intestine group, Treating lung and large intestine group, each groups has 10 rats. The STC model of Model control group and the three treatment groups was established by DC garage.While Blank control group gave the same dose of 0.9% saline. When the model was established, three treatment groups were fed with Chinese herbal compound which corresponds with it. The period of treatment was 14 days. While Blank control group and Model control group gave the same dose of 0.9% saline. After the treatment, Use Activated charcoal propulsion experiment to test the intestinal transmission function of five groups of rats. Observe the pathological change of lungs and colon by pathological section. Expression of VIP, SPand5-HT in lungs and colon tissue was detected with Immunocytochemistry. Results:1.Activated charcoal propulsion experiment after the model was established:The first black stool discharge time of Blank control group was less than the rest of four groups (P<0.05),24h stool number and 24h stool weight were more than the rest of four groups (P<0.05), difference was statistically significant.2.Activated charcoal propulsion experiment after the treatment of TCM:The first black stool discharge time of Model control group and Treating lung group was more than the rest of three groups (P<0.05),24h stool number and 24h stool weight were less than the rest of three groups(P<0.05), difference was statistically significant. Model control group compared with Treating lung group (P<0.05), difference was no statistically significant. Comparison of the remaining three groups (P<0.05), difference was no statistically significant.3.The pathological changes of lungs and colon tissue:Blank control group:Structural integrity, no hyperemia and inflammatory cells invasion in lungs and colon tissue. Model control group: Only had few inflammatory cells around the alveoli and the capillaries. The colonic mucosa, serosa and muscularis invasion of inflammatory cells. A small amount of vacuoles degeneration in the myenteric neurons. Treating lung group: Structural integrity, no hyperemia and inflammatory cells invasion in lung. The colonic mucosa and serosa invasion of few inflammatory cells. Myenteric neuron was more than the Model control group. Treating large intestine group: Only had few inflammatory cells around the capillaries. Myenteri neurons were more than the Treating lung group. No hyperemia, edema and inflammatory cells invasion. Treating lung and large intestine group:Structural integrity,no hyperemia and inflammatory cells invasion in lungs and colon.4. Immunohistochemistry results of lungs and colon tissue: ①VIP in lungs:Model control group was less than the rest of four groups (P<0.05), difference was statistically significant. Treating large intestine group was less than the Blank control group, Treating lung group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating lung group was less than the Blank control group and Treating lung and large intestine group (P<0.05), difference was statistically significant. Blank control group compared with Treating lung and large intestine group (P<0.05), difference was no statistically significant.②VIP in colon:Model control group was more than the rest of four groups (P<0.05), difference was statistically significant. Treating lung group was more than the Blank control group, Treating large intestine group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating large intestine group was more than Treating lung and large intestine group (P<0.05), difference was statistically significant. Blank control group compared with Treating large intestine group and Treating lung and large intestine group (P<0.05), difference was no statistically significant. ③SP in lungs:Model control group was more than the rest of four groups (P<0.05), difference was statistically significant. Treating large intestine group was more than the Blank control group, Treating lung group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating lung group was more than the Blank control group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Blank control group compared with Treating lung and large intestine group (P<0.05), difference was no statistically significant. ④SP in colon:Model control group was less than the rest of four groups (P<0.05), difference was statistically significant. Treating lung group was less than the Blank control group, Treating large intestine group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating large intestine group was less than Treating lung and large intestine group (P<0.05), difference was statistically significant. Blank control group compared with Treating large intestine group and Treating lung and large intestine group (P<0.05), difference was no statistically significant. ⑤5-HT in lungs:Model control group was more than the rest of four groups (P<0.05), difference was statistically significant. Treating large intestine group was more than the Blank control group, Treating lung group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating lung group was more than the Blank control group and Treating lung and large intestine group (P<0.05), difference was statistically significant. Blank control group compared with Treating lung and large intestine group (P<0.05), difference was no statistically significant. ⑥5-HT in colon:Model control group was less than the rest of four groups (P<0.05), difference was statistically significant. Treating lung group was less than the Blank control group, Treating large intestine group and Treating lung and large intestine group(P<0.05), difference was statistically significant. Treating large intestine group was less than Treating lung and large intestine group(P<0.05), difference was statistically significant. Blank control group compared with Treating large intestine group and Treating lung and large intestine group (P<0.05), difference was no statistically significant. ⑦The correlation analysis of VIP, SP and 5-HT in lungs and colon:VIP in lungs was positively correlated with VIP in colon of Blank control group, VIP in lungs was negatively correlated with VIP in colon of the rest of four groups. SP in lung was no correlated with SP in colon of Blank control group, SP in lungs was negatively correlated with SP in colon of the rest of four groups.5-HT in lungs was positively correlated with 5-HT in colon of Blank control group, 5-HT in lungs was negatively correlated with 5-HT in colon of the rest of four groups. Conclusion:1.Experiments have shown that STC can cause the pathological changes of lung tissue by influencing on the secretion or expression of VIP, SP and 5-HT in the lungs of rats.2. Treating lung method (Bu-fei soup) can recover the pathological changes of lung tissue, and has certain improvement effect to the pathological changes of colon tissue and the function of ENS of rats at the same time; The mechanism is Treating lung method can adjusting the secretion of VIP, SP and 5-HT in the lungs, and redistribute it’s in the lungs and colon and exert its biological activities.3. Treating large intestine method (Ma-ren pills) can recover the colon transit function of STC rats, and has certain improvement effect to the pathological changes of lung tissue caused by STC at the same time; The mechanism is Treating large intestine method can adjusting the secretion of VIP, SP and 5-HT in the colon, and redistribute it’s in the lungs and colon and exert its biological activities.4. Treating lung and large intestine simultaneously (Bu-fei soup and Ma-ren pills) can recover the pathological changes of lung and colon tissue, and recover the colon transit function of STC rats. The mechanism is Treating lung and large intestine simultaneously can recover the pathological changes of lungs and colon tissue, and recover the ENS function of STC rats by adjusting the secretion and redistribute of VIP, SP and 5-HT in the lungs and colon at the same time.5. Experiments prove that one of the mechanism of treating STC with Treating lung and large intestine simultaneously (Bu-fei soup and Ma-ren pills) is that VIP, SP and 5-HT can connect with each other in lung and large intestine. VIP, SP and 5-HT was the material which contact with lungs and large intestine, is one of the material basis in the theory of The lung and the large intestine being interior-exteriorly related".
