Objective:In this experimental study,the effects of different acupuncture pretreatments on the effects of Neiguan points on paroxysmal atrial fibrillation rats were compared by animal experiment.The duration of atrial fibrillation and the amplitude of P-wave of atrial repolarization parameters were recorded by ECG.And the variation of P wave dispersion as a therapeutic effect judgment index,comparing the different acupuncture depths on the pathological structure changes of right atrial myocytes in paroxysmal atrial fibrillation rats to compare the effects of different acupuncture depths on the curative effect,aiming at acupuncture clinical treatment of atrial fibrillation(Atrial fibrillation)provides experimental evidence.Methods:Forty male Wistar rats were randomly divided into normal group,model group,shallow acupuncture group and deep acupuncture group,with 4 groups,10 in each group.In the normal group,saline was injected into the dorsal vein of the rat penis.In the model group and the pretreatment group,a mixture of Ach 66μg and CaCl 210 mg was injected into the dorsal penile vein of the rat to induce paroxysmal atrial fibrillation,and continuous administration was performed.The model was made in 7d,and the duration of atrial fibrillation was recorded with an electrocardiogram.In the shallow acupuncture group,the needle was obliquely inserted into the Neiguan point in a direction inclined by 15 degrees,and the needle tip was inserted into the epidermis;the deep acupuncture group was pierced into the Neiguan point by a straight puncture method,and after the needle was left for 20 minutes,Injection molding drugs.The duration of atrial fibrillation was measured by electrocardiogram for 7 days of continuous administration.The general condition of each group of mice,such as hair status,defecation,and mental status,was observed.The right atrial tissue of rats was taken and stained and observed by light microscopy.The changes of right atrial tissue structure in each group were compared.The duration of atrial fibrillation after treatment,the changes of P-wave width and P-wave dispersion of atrial repolarization related parameters of electrocardiogram were calculated as the evaluation indexes.Statistical analysis was performed using IBM SPSS 20.0 statistical software.Results:1.General situationMental state;the normal group of rats was positive and responsive;the model group and the shallow acupuncture group showed poor general condition,exhausted,tired,lying,shortness of breath,purple claws and lips,and large group with the model group Compared with the rats,the rats in the deep acupuncture group were relatively more sensitive,and the lips were not purple.Fur color:the normal group hair is white and bright;the model group hair is shed and the color is yellow and dark;the acupuncture pretreatment group is improved in the later stage,and the color is improved.2.Duration of atrial fibrillationAF duration of the model group was gradually prolonged,and there was a statistically significant difference compared with the normal group(P<0.01).On the 7th day,there was significant difference between the deep acupuncture pretreatment group and the model group(P<0.05).There was no significant difference between the shallow acupuncture pretreatment group and the model group on the seventh day(P>0.05).3.P wave width comparison analysisAfter 7 days of injection molding,the P wave width of the four groups of rats changed.The difference between the deep acupuncture pretreatment group and the model group was statistically significant(P<0.01)5 and the statistical analysis had extremely significant differences(P<0.05).There was no significant difference between the shallow acupuncture pretreatment group and the model group(P>0.05).4.Comparative analysis of P wave dispersionAfter 7 days of injection molding,the P wave dispersions of the four groups were all changed,and the variance analysis(P<0.01)showed significant difference.The deep acupuncture pretreatment group was lower than the model group.There was a statistical difference(P<0.05).There was no significant difference in P wave dispersion between the shallow acupuncture pretreatment group and the model group(P>0.05).5.HE(hematoxylin-eosin staining)staining resultAfter HE staining,the myocardial cells in the model group were disordered under high magnification,the muscle fibers were loosely arranged,the continuous myocardial fibers were partially broken and accompanied by focal muscle fiber fusion.The epicardium of the normal group was smooth,and the subecular myocardial fiber structure was clear.Occasionally,myocardial rupture and eosinophilic changes were observed in some areas of the deep acupuncture pretreatment group,and no obvious changes were observed in the endocardium.In the shallow acupuncture preconditioning group,the epicardium was smooth,the structure of the subepithelial myocardial fibers was clear,the eosinophilic changes and myocardial rupture were observed in the whole myocardium,the local muscle gap was significantly widened,no obvious inflammatory changes were observed,and the endocardium was partially visible.Focal necrotic changes were relatively heavier than other treatment groups.Conclusion:1.The rat model of paroxysmal atrial fibrillation was induced by continuous injection of calcium chloride-acetylcholine mixed solution in the dorsal vein of the penis for 7 days.The electrocardiogram showed that the typical atrial fibrillation electrocardiogram showed successful modeling,compared with the rat tail vein injection.The penile vein injection is easier to operate and reproducible,and is suitable for research and application in basic animal experiments.2.Deep acupuncture pretreatment Neiguan acupoint can shorten the atrial repolarization parameter wave width and wave-disturbing degree of atrial fibrillation in rats and the number of atrial fibrillation in humans,so that the atrial repolarization process tends to be normal.3.Deep acupuncture pretreatment in Neiguan acupoint can alleviate the damage of atrial fibrillation on rat atrial myocytes,protect the heart structure,improve atrial remodeling caused by atrial fibrillation,and reduce the mortality of rats.4.Deep acupuncture preconditioning can reduce the duration of atrial fibrillation,can effectively fight atrial fibrillation,shorten the duration of atrial fibrillation,and is better than the shallow acupuncture group.
