ObjectiveTo observe the effects of ampk-acc signaling pathway on glucose and lipid metabolism by interfering with ZDF rats with Polygonatum Odoratum polysaccharide.Through this method to explore its mechanism for clinical basis.MethodsA randomized controlled trial design was used in this study.Diabetes was induced in 40 male ZDF rats aged 7-8 weeks with high-fat diet,and 10 ZLF normal rats were fed with normal diet as the control group.After 4 weeks,the induced diabetes rats were selected and randomly divided into normal control group,metformin group,low dose of polysaccharide of rhizoma cyperi and high dose of polysaccharide of rhizoma cyperi.Metformin group was given metformin 150mg/kg/day.The PoP low dose group was given 1.26g/kg/day.The PoP high dose group group was given 2.52g/kg/day.The control group was given distilled water of the same volume by gavage.In total,eight weeks.Fasting glucose(FBG)was monitored at 2,4,and 6 weeks.Oral glucose tolerance test(OGTT)was performed at 7 weeks.The rats were anesthetized with pentobarbital sodium at week 8.The levels of glycosylated serum protein(GSP),serum lipid which including total cholesterol(TC),triglyceride(TG),LDL cholesterol(LDL-C)and free fatty acid(FFA)were measured by automatic blood biochemical analyzer.The weight of the rat liver was weighed,and the liver tissues were collected.The expressions of AMPK 1,AMPK2 and phosphorylated acetylcoa carboxylase(P-ACC)protein in liver tissues were detected by western-blot.ResultsCompared with the normal group,FBG was significantly increased in the model group at 2,4 and 6 weeks,while FGB,ogtt-auc and GSP were significantly increased in the model group(P<0.05,P<0.01),TG,CHO,LDL-C and FFA were significantly increased in the model group(P<0.01).Histological observation showed that the liver cells of ZDF rats in the model group showed morphological destruction and obvious fat deposition by HE staining in liver sections.Western protein assays showed that liver AMPK 1 and AMPK 2 protein expressions were significantly increased,and p-acc protein expression was significantly increased(p<0.05).Compared with the model group,FGB,OGTT-AUC and GSP in the metformin group,PoP high group and PoP low groups were significantly reduced(P<0.05).TG,CHO,LDL-C and FFA were significantly decreased in the metformin group and the PoP high group(P<0.05),but only FFA was significantly decreased in the PoP low group(P<0.05),and only FFA was decreased in other indexes(no statistical difference).The liver cells in the drug administration group were all orderly arranged and the fat deposition was significantly reduced compared with the model group.Compared with the model group,the expressions of AMPK 1 and AMPK 2 protein in liver and P-ACC protein were significantly increased in the metformin group and the PoP high group rats' liver(p<0.05).The expression of AMPK 1 and P-ACC protein increased significantly in PoP low group(p<0.05),while the expression of AMPK 2 protein increased without statistical difference.Conclusion1.Polygonatum Odoratum polysaccharide can reduce FBG,OGTT and GSP of ZDF rats with type 2 diabetes mellitus and dyslipidemia,so as to regulate them glucose metabolism.2.High dose of Polygonatum Odoratum polysaccharide can reduce the blood lipids of ZDF rats with type 2diabetes mellitus and dyslipidemia,and effectively improve the liver fat deposition in diabetic rats,so as to regulate the abnormal lipid metabolism.3.Polygonatum Odoratum polysaccharide can increase the expression of AMPK 1 and AMPK 2 protein in the liver of ZDF rats with type 2 diabetes mellitus and dysplasia,thus promoting the phosphorylation of ACC protein.Thus,it can reduce the lipid deposition of liver cells,reduce liver steatosis,improve insulin sensitivity.Finally the disorder of glucose and lipid metabolism can be improved.
