OBJECTIVE:To observe the effect of Bajitian on type ? collagen in KOA joints of rats,and to detect the expression of IL-1,TNF-alpha,MMP3 and MMP13 in serum of rats,and to detect the expression of type ? collagen in synovium and cartilage of joints.Finally,the results were analyzed.METHODS:Thirty male rats were fed for 7 days and randomly divided into five groups:blank control group(K group),model control group(S group),low concentration group(BL),medium concentration group(BM)and high concentration group(BH)of Morinda officinalis.Except group K,all the models were induced by iodoacetic acid.Rats were immobilized in supine position after intraperitoneal anesthesia with hydric aldehyde.The hair around the knee joint was removed with a scalpel to expose the knee joint completely.After 3 times of iodophor disinfection,the knee joint of slightly bent rats was injected with 0.1 mL iodoacetic acid solution(1 mg/mL)into the right knee joint cavity.After the injection,the knee joint was gently bent several times to make the iodoacetic acid solution fully infiltrate the joint cavity.The contralateral side served as blank control.Later to determine the success of the model.After the model was established,the rats were fed for 2 weeks,waiting for the inflammation to form.The circumference of the knee joint was recorded with vernier caliper every day after the establishment of the model.Compared with group K,the circumference of the knee joint of the four groups of rats was significantly increased and the activity of the rats was decreased after 1-2 days of the establishment of the model.Two weeks later,one rat was randomly selected from each group to dissect the knee joints.The right knee joint cartilage was obviously rough,soft tissue adhesion was obvious,and ulceration was seen in some synovium,which indicated that the model was successful.Two weeks later,the blank control group(group K)was fed with conventional diet without any intervention.The control group(group S)was given normal saline once a day,and the low,medium and high concentration groups were given Morinda officinalis 4g/Kg,8 g/Kg,16 g/Kg intervention respectively.Morinda officinalis was given once a day.The dosage and concentration of Morinda officinalis were calculated by body surface area between human and animal.The equivalent dose of rats was 6.3 times that of normal people.After 4 weeks of drug intervention,all five groups of rats were anesthetized with 5%chloral hydrate,then sacrificed after taking abdominal aortic blood in vacuum.Blood samples were centrifuged in centrifuge,and serum samples were collected.The expression of IL-1beta and TNF-alpha protein was analyzed by ELISA,and the expression of MMP-3 and MMP-13 protein was analyzed by ELISA.The right knee joint of rats was incised uniformly.The synovium and articular cartilage were carefully stripped and placed in the cryopreservation tube respectively.The cryopreservation tube was placed in the refrigerator at-80 C in time.The expression of type ? collagen in synovium and cartilage was detected by immunohistochemistry.RESULTS:Based on the results of cytokine test,the serum concentration of IL-1beta in the low,medium and high concentration groups of Bajitian was lower than that in the model control group(P<0.01).In addition,the serum concentrations of TNF-a,MMP3 and MMP13 in different concentration groups of Bajitian were significantly lower than those in model control group(P<0.01).The expression of Col-? in synovium and cartilage was detected by immunohistochemistry.The expression of Col-? in different concentration groups of Bajitian was higher than that in model group(P<0.05).CONCLUSION:We can prove that Bajitian can promote the expression of Col-? in KOA joints,thus protecting articular cartilage and alleviating inflammation.Moreover,Bajitian L.has significant inhibitory effects on cytokines such as IL-1beta,TNF-alpha and matrix metalloproteinase in the treatment of KOA j oints.Therefore,through this experiment,we can find that Bajitian can protect chondrocyte,eliminate inflammatory reaction and alleviate clinical symptoms by inhibiting the secretion of proinflammatory factors and matrix metalloproteinase,and promoting the expression of Col-?.
