Lindera aggregate?Sims?Kosterm.is a traditional Chinese medicine in China and also one of the major medicinal materials in Hunan province.Its processed have being widely used in clinical practice in recent years.However,there were few reports on the study about quality control and evaluation of the effects of vinegar processed Lindera aggregate on“xingqi”.Therefore,in this paper,we took vinegar processed L.aggregate as the research object,set up fingerprint of each fractions for defining quality control standards,basd on Effects of Gastric Emptying and Gastrointestinal Hormones in on stomach cold models in rats to decrease adverse effects and ensure security in clinic.The main research contents are as follows:1 Fingerprint and pattern recognition of vinegar processed Linderae Radix from different habitatsObjective:To establish the fingerprint of vinegar processed Linderae Radix from different habitats with HPLC method,and to evaluate the quality through the similarity calculation and chemical pattern recognition.Methods:The separation was performed on an Agilent 5 HC-C18?2??250 mm×4.6 mm,5?m?at?25±5??.A binary gradient elution system was composed of methanol and water containing 0.5%formic acid and the flow rate was 1.0 mL·min-1.The detection wavelength was 235 nm.Similarities of fingerprints of 53batches of crude and vinegar processed Linderae Radix were analyzed by software Similarity Evaluation System for Chromatographic Fingerprint of TCMs?Version2012A?.Then principal component analysis?PCA?and orthogonal partial least squares discriminate analysis?OPLS-DA?were performed with SIMCA 13.0.Results:The HPLC fingerprints of 53 batches of crude and vinegar processed Linderae Radix were established successfully in this study.The similarities of crude and vinegar processed Linderae Radix were between 0.51 and 0.70,indicating the difference between the crude and processed samples.All the samples were classified into two categories by PCA.The OPLS-DA result showed that the primary differentiating components were norisoboldine,linderalactone and linderane.Conclusion:The proposed method provided basis for quality control of and vinegar processed Linderae Radix.And chemical pattern recognition is proved to be helpful in comprehensive quality control and evaluation of Linderae Radix before and after the process.2 Effects of vinegar Lindera on stomach cold models in ratsObjective:To investigate the effects of vinegar Lindera on gastric emptying,small intestinal propulsion rate and motilin?MTL?,gastrin?GAS?,Cyclic adenosine monophosphate?cAMP?,Cyclic guanosine monophosphate?cGMP?and cAMP·cGMP-1 of plasma in rat stomach cold models.Methods:Totally 80 SD rats were randomly divided into blank group?n=10?and model group?n=70?;Rats were soaked with 0?water 20 min,the depth of the water is 5 cm,lavaged immediately in ice water to 20 ml·kg-1 in modeling group,twice a day,for 7 day.After successfully establishing the models,rats were randomly divided into model group,Lindera group,vinegar Lindera low,moderate,high dose group and dupanlitazone group with 10 rats in each group.Administration groups were given relevant medicine once a day,3day.Gastric emptying rate and small intestinal propulsion rate of rats in each group were measured by phenol red content method.The contents of motilin?MTL?,gastrin?GAS?,Cyclic adenosine monophosphate?cAMP?,Cyclic guanosine monophosphate?cGMP?in serum were determined by ELISA method.Results:Compared with blank group,gastric emptying rate and small intestinal propulsion rate of rats were decreased significantly in model group,and the serum contents of MTL and GAS were decreased significantly,while the content of cAMP·cGMP-1was increased significantly?P<0.01?.Compared with Lindera group,the gastric emptying rate and small intestinal propulsion rate of vinegar Lindera group were increased significantly,and the serum contents of MTL and GAS were increased significantly,while the content of cAMP·cGMP-1 was decreased significantly?P<0.05?.Conclusion:Vinegar Lindera aggregate can accelerate gastric emptying and small intestinal propulsion rate,increase serum contents of MTL,GAS and cAMP·cGMP-1.
