Objective:To observe the expression of superoxide dismutase(SOD)and malondialdehyde(MDA)in renal tissue of rats with unilateral ureteral obstruction,and to investigate the effect of butylphthalide on oxidative stress in rats with renal interstitial fibrosis.Methods: Select clean level 54(male)in rats,were randomly divided into 3 groups:Sham-operated group(group A),model group(group B),treatment group(group C)after anesthesia open abdominal separation of the left ureter.The group B with group C and in close proximity to renal pelvis left ureteral ligation;group C espectively for butylphthalide180 mg/kg/d lavage;group B give equal saline lavage;the group A only isolated the ureter,no ligation give equal saline lavage too.Six rats in each group were randomly selected and sacrificed on 2,3 and 7 days after surgery.Serum samples and kidney tissues were taken,serum levels of blood urea nitrogen(BUN)and serum creatinine(Scr)were detected,and SOD activity was determined by Xanthine oxidase method,MDA by Thiobarbituric acid method,and pathological changes of the kidney were observed by HE and Masson staining.Results: BUN,Scr and the expression of MDA in group C were higher than those in group A,but significantly lower than group B,and the expression of SOD in Group C were higher than group B(P<0.05).Pathological changes of the kidney in group C were significantly decreased compared with group B,but still more serious than those in group A.Conclusion:(1)Oxidative stress is involved in the development of renal interstitial fibrosis.(2)Butylphthalide plays a significant role in the regulation of renal interstitial fibrosis in rats in monolateral ureteral obstruction,and the reasons for this may be related to the regulation of the activity of SOD and MDA.
