Purposes:To observe the effect of "He-wei-li-qi Decoction" in rats with functional dyspepsia of liver qi stagnation type on behavior,gastrointestinal motility and c-kit / SCF(dry cell factor)signaling pathways,to explore the possible mechanism of He-wei-li-qi decoction for treating functional dyspepsia from the direction of gastrointestinal dysfunction,providing a new theoretical basis for traditional Chinese medicine to treatmentfunctionaldyspepsia.Material and methods:40 healthy Wistar SPF rats were selected after after the open field test score,they were randomly divided into 4 groups: blank group,model group,and stomach qi gas group and domperidone group though random number table method,with 10 in each group.The blank group of 10 rats were without any treatment,giving normal breeding and away from modeling environment,the remaining three groups of rats were using tailgating stimulation + irregular feeding composite method to establish liver qi stagnation functional dyspepsia model,the time was 14 days.During the treatment period,rats in the blank group and model group were not given any treatment;giving the gavage treatment by the solution of He-wei-li-qi decoction to the He-wei-li-qi decoctiongroup,2 times a day for 14 days;giving the gavage treatment of domperidone solution to domperidone group,2 times a day for 14 days.The weight,sugar preference test and open-field test of rats were measured on the time of 1st day before the experiment,the 14 th day in the experiment and the 28 th day in the experiment.At the end of the experiment,the rats were anesthetized and sacrificed,and the gastric emptying rate and intestinal propulsion were measured to determine the indexes of gastric emptying rate and intestinal propulsion rate;immunohistochemistry was used to detect the expression of c-kit and SCF in gastric antrum and small intestine;western-blot was used to detect the expression of c-kit protein and SCF protein in gastric antrum and small intestinal mucosa;RT-PCR was used to detect the expression of c-kit mRNA and SCF mRNA in gastric antrum and small intestine;observing the effect of he-wei-li-qi decoction on c-kit protein,SCF protein,c-kit mRNA and SCF mRNA in gastric antrum and small intestinal mucosa of rats with liver-qi stagnation dysfunction model.Results: 1.One day before the start of the experiment,there was no significant difference in body weight and behavioral indexes among the four groups,with no significant difference(P>0.05).After modeling,the general status of rats in blank group is still basically the same as before modeling,while the three other groups of rats showed disorderly hair disorderly yellow hair,stool foul,or even loose stools;active significantly reduced compared with the previous,lazy and often tired and slow weight growth;the preference rate of syrup decreased significantly;the level of open box test and the score of vertical movement decreased significantly.The difference between each model group and blank group was significant(P<0.05).After the treatment,compared with the model group,the hair of the rats in the he-wei-li-qi decoction group and the domperidone group gradually softened and lightened,the stool gradually formed,the spirit improved,the mood gradually became stable and calm,the amount of activity increased,and the feed intake increased;weight,open box experimental level and vertical scores were significantly higher than the model group(P<0.05).2.Compared with the blank group,the gastric residual rate in the model group was significantly increased,and the small intestine propulsion rate was significantly decreased,the differences were statistically significant(P<0.05).After intervention,the gastric residual gastritis and domperidone could obviously reduce the gastric residual rate and promote the intestinal propulsion rate(P<0.05).3.Compared with the blank group,the expression of c-kit and SCF in gastric antrum and intestinal mucosa in model group were significantly lower than those in blank control group(P<0.05)by immunohistochemistry.Compared with the model group,the expressions of c-kit and SCF in the gastric antrum and the intestinal mucosa in the he-wei-li-qi decoction group and the domperidone group were significantly higher than those in the model group(P<0.05).Compared with domperidone group,there was no significant difference with the he-wei-li-qi decoction group(P> 0.05).4.Compared with the blank group,the expression of c-kit protein and SCF protein in gastric antrum and intestinal mucosa in model group were significantly lower than those in blank control group by Western Blot(P<0.05).Compared with the model group,the expressions of c-kit and SCF in the gastric antrum and the intestinal mucosa in the he-wei-li-qi decoction group and the domperidone group were significantly higher than those in the model group(P<0.05).Compared with domperidone group,there was no significant difference with the he-wei-li-qi decoction group(P> 0.05).5.Compared with the blank group,the expression of c-kit mRNA and SCF mRNA in gastric antrum and intestinal mucosa in model group were significantly lower than those in blank control group by Western Blot(P<0.05).Compared with the model group,the expressions of c-kit mRNA and SCF mRNA in the gastric antrum and the intestinal mucosa in the he-wei-li-qi decoction group and the domperidone group were significantly higher than those in the model group(P<0.05).Compared with domperidone group,there was no significant difference with the he-wei-li-qi decoction group(P> 0.05).Conclusion: 1.Though the detection of animal weight,behavioral observation and gastric motility test,it proved that he-wei-li-qi decoction can effectively relieve liver qi stagnation type FD rats mental and behavioral discomfort symptoms.2.He-wei-li-qi decoction can improve gastrointestinal dysfunction in liver qi stagnation type FD rats,and the mechanism may be related to enhancing the expression of c-kit and SCF,activating the c-kit / SCF signaling pathway,thereby promoting gastrointestinal motility.
