Objective: To investigate the effect of Tangshen’an decoction on Nrf2/ARE pathway in renal tissue of experimental type 2 diabetic rats and to study its protective effects on the kidney.Materials and Methods: Fifty healthy male SD rats were randomly divided into normal group and model building group.Rats in model group were fed with high fat and high glucose.Type 2 diabetes model was established by intraperitoneal injection of streptozotocin(STZ)at a dosage of 35mg·kg-1.Rat models were randomly divided into model group,Tangshen’an low,medium and high dose group and western medicine group.Each treatment group was given the appropriate treatment.After 8 weeks,the general situation in rats was observed.24 h urine microalbumin was detected by collecting urine sample.Fasting blood glucose plasma lipid and renal function were measured by abdominal aorta blood sampling.The kidneys were collected and the expression of Nrf2 and HO-1 protein in kidney were detected by immunohistochemistry.The protein and m RNA expression of NF-κBp65 in kidney were detected by Western blot and RT-PCR.Result:1.General situation of rats in each group: The rats in the model group showed diminished activity,bleak hair color,more drink,more food,sluggishness,unresponsiveness and even bowstring curling body.After 8 weeks,the body weight of the model group and each treatment groups was lower than that of the normal group(P<0.05).2.Changes of Fasting Blood Glucose in rats in each group:After 8 weeks,fasting blood-glucose of the model group and each treatment group was significantly higher than the normal group(P<0.01).3.Changes of blood lipids in rats in each group:CHOL in model group were higher than that in normal group(P<0.05);TG in the model group,Tangshen’an low,medium dose group was higher than the normal group(P<0.01,P<0.05).TG in each treatment group was lower than that in model group(P<0.01,P<0.05).LDL-C in model group,Tangshen’an low-dose group and western medicine group were significantly higher than those in normal group(P<0.05),LDL-C in model group were higher than that in Tang shen’an high-dose group(P<0.05).4.Changes of renal function in rats in each group:The BUN in model group and western medicine group was higher than that in normal group(P<0.01,P <0.05).The BUN in middle and high dose Tangshen’an group was lower than that in model group(P<0.01,P<0.05).The Scr in the model group and the low dose of Tangshen’an group were significantly higher than that in the normal group(P<0.01).The Scr in middle and high dose Tangshen’an group and western medicine group was lower than that in the model group(P<0.01,P<0.05);.5.Changes of 24-hour urine microalbumin in rats in each group:24h urinary album inin the model group and each treatment group was higher than that in the normal group(P<0.01).6.Changes of renal tissue Nrf2 in rats in each group:The expression of Nrf2 protein in renal tissue of model group and each treatment group was higher than that of normal group(P<0.01),and the expression of Nrf2 protein in middle and high dose group and western medicine group was higher than that in model group(P<0.01).The expression of Nrf2 protein in renal tissue of the low dose of Tangshen’an group was lower than that of western medicine group(P<0.01).7.Changes of renal tissue HO-1 in rats in each group:The expression of HO-1 in model group and each treatment group was higher than that in normal group(P<0.01)andthe expression of HO-1protein in middle and high dose group and western medicine group was higher than that in model group(P<0.01).The expression of HO-1 in the low dose Tangshen’an group was lower than that in the western medicine group(P<0.01)and theexpression of HO-1in high dose Tangshen’an group was higher than that in western medicine group(P<0.05).8.The expression of renal tissue NF-κBp65 in rats in each group:8.1Western blot was used to detect the expression of NF-κBp65 protein in rat kidney:The expression of NF-κBp65 in renal tissue of model group and each treatment group was higher than that of normal group(P<0.01,P<0.05).The protein expression of NF-κBp65 in renal tissue of each treatment group was lower than that in model group(P<0.01).The expression of NF-κBp65 in renal tissue of low dose Tang shen’an group was higher than that in western medicine group(P <0.01).8.2The expression of NF-κBp65m RNA in rat kidney tissue was detected by RTPCR:The expression of NF-κBp65 m RNA in renal tissue of model group,low and middle dose Tangshen’an group and western medicine group were higher than that in normal group(P<0.01).The expression of NF-κBp65 m RNA in kidney of low dose Tangshen’an group was higher than that in western medicine group(P<0.01).Conclusion:1.Tangshen an decoction can reduce fasting blood glucose,24h urinary albumin,regulate blood lipids,protect renal function in diabetic rats.2.Tangshen’an decoction activates Nrf2/ARE signaling pathway,up-regulates the expression of the antioxidant enzyme gene HO-1,inhibits the expression of NF-κBp65 m RNA and protein,improves the renal inflammatory response and exerts the anti-oxidativeeffect.
