The Effect Of Yukui Fang On SOD, MDA, NO, INOS And MPO In Experimental Ulcerative Colitis Rats

Purpose:To observe the effect of Yukui Decoction on the expression of serum superoxide dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO),and the content of colon tissue myeloperoxidase(MPO),inducible nitric oxide(iNOS)of the experimental ulcerative colitis(UC)rats,and to explore the possible mechanism of Yukui Decoction in treating ulcerative colitis,provide experimental basis for the clinical application.Material and method:70 healthy male SD rats were randomly divided into blank group(n=8)and molding group(n=62),The rats in the molding group were established UC model by using the three nitrobenzene sulfonic acid TNBS-ethanol method.On the next day after successful modeling,the molding rats were randomly divided into model group,YuKui Decoction small,medium and large dose group and mesalazine group.The blank group and the model group were given the same dose of 0.5% sodium carboxymethylcellulose and each treatment group was given different dosage of YuKui Decoction2.835g/kg,5.67g/kg,11.34g/kg and mesalazine 0.36g/kg for 14 days.After the last treatment,all rats fasting 24 hours then kill all rats.under the microscope observed the pathological morphology of the colonic mucosa,xanthine oxidase detected of SOD activity,thiobarbituric acid colorimetry detected the content of MDA,nitrate reductase detected the level of NO,immunohistochemistry detected the activity of iNOS,and content of MPO was detected by spectrophotometer colorimetry.Results:1.General observation and DAI scoring results:Molding group rats showed mental depression,poor reaction,unclean fur,reduced food intake,weight loss,incomplete stool and perianal contamination.After treatment,except the model group,the symptoms of the other groups improved,but compared with the blank group,the recovery of the above symptoms was slightly worse.The results of DAI scoring showed that each dosage of YuKui Decoction groups and mesalazine group were lower than model group,and the difference was statistically significant(P < 0.05);there was no significant difference compare differentdosage of YuKui Decoction groups with mesalazine group(P > 0.05).2.Pathological and histological changes:Colonic mucosa of blank group rats was smooth surface,without congestion and edema,and glands arranged regularly;model group rats was erosion,congestion and edema,with extensive ulceration of mucosa,infiltration of inflammatory cells;in the four treatment groups mucosal congestion and edema improved markedly,and inflammatory cells decreased,goblet cells increased,and the degree of ulcer was in recovery stage.Compared with the blank group,model group rats CMDI and TDI scores increased significantly(P<0.05),compared with the model group treatment groups showed a downward trend(P<0.05)There was no significant difference between different dosage of YuKui Decoction groups and mesalazine group(P>0.05).3.Indicator detection:Model group serum SOD activity was significantly lower than blank group(P < 0.05),serum MDA,NO levels in model group were higher than blank group(P <0.05),the content of colon tissue MPO,iNOS were higher than those in blank group(P <0.05).After treatment with Yukui Decoction,serum SOD activity increased,serum MDA,NO and tissuesMPO,iNOS content decreased significantly,which had statistical significance compared with model group(P < 0.05).Yukui Decoction large dose group and Mesalazine group had better therapeutic effect,There was no significant difference between the two groups(P > 0.05),followed by Yukui Decoction medium dose group and Yukui Decoction small dose group.Conclusion:Yukui Decoction can improve the general condition of UC rats.Yukui Decoction can reduce the expression of NO and iNOS by increasing the activity of SOD,decreasing the content of MDA and MPO,and clearing the excess free radicals,thus reducing the damage of colonic tissue and promoting the healing of mucosa.
