Purpose: In this study,we established a mouse model of mild cognitive impairment caused by atherosclerosis,and observed the intervention effect of Erchen Decoction and Taohong Siwu Decoction combined with acupuncture on model mice.According to the changes of acetylcholinesterase(ACh E),acetylcholine transferase(Ch AT),interleukin-1?(IL-1?),tumor necrosis factor alpha(TNF-?)and amyloid ?-protein(A?)in model mice,the mechanism of Erchen Decoction and Taohong Siwu Decoction combined with acupuncture on the cholinergic system and inflammatory response of the model mice was explored.Based on the theory of“cardio-cerebral disease”,this study explored the effect and mechanism of atherosclerosis on cognitive function,aiming to provide experimental basis and theoretical guidance for clinical prevention and treatment of mild cognitive impairment caused by atherosclerosis.Material and method:10 C57BL/6J mice were selected as blank control group,and fed with normal mouse feed.40 Apo E-/-mice were selected as the model control group,and fed with high-fat diet feed.After 12 weeks of feeding,the blood lipid level of mice was detected by automatic biochemical analyzer,the pathological morphology of mice arterial was detected by HE staining,and the cognitive ability of mice was observed by Morris Water Maze,among which the learning ability of mice was detected by place navigation experiment and the memory ability of mice was detected by spatial probe experiment.After confirming the successful establishment of the model,mice in the blank control group were still treated as blank group.Mice in the model control group were randomly divided into model group,traditional Chinese medicine group(TCM group),acupuncture group and the combination of acupuncture and traditional Chinese medicine group(combination group).The TCM group was given intragastric administration of Erchen Decoction and Taohong Siwu Decoction,0.2m L/time,1 time/d.The acupuncture group selected Baihui,Guanyuan and bilateral Fenglong,electroacupuncture was kept for 15 min,the left and right sides were performed alternately,1 time/d.The combination group was given the treatment of TCM group and acupuncture group at the same time.The blank group and the model group were given the same degree of grasping and fixation,and given intragastric administration of distilled water,0.2m L/time,1 time/d.All treatment methods last for 28 days,during the treatment period,mice in each group were fed continuously according to the established feeding mode.At the end of treatment,behavioral testing detected by Morris water maze,the expression of Ch AT and ACh E was detected by immunohistochemistry,and the contents of IL-1?,TNF-? and A? were determined by enzyme-linked immunosorbent.Results:1.After modeling,the results of blood lipid level test showed a significant increase in the serum levels of TC,TG,and LDL in the model control group(P<0.01),with a significant decrease in HDL level(P<0.01),compared with the blank control group.2.After modeling,HE staining test results showed larger arterial plaques in the model control group,in contrast with the blank control group.3.After modeling,Morris water maze test results show that,in the place navigation experiment,with the increase of training time,the escape latency of each group of mice decreased.Compared with the blank control group,the escape latency and the swimming distance of mice in the model control group were obvious increased(P<0.05).In the spatial probe experiment,compared with the blank control group,the frequency of crossing the platform in the model control group was obvious reduced(P<0.05).4.After treatment,Morris water maze test results showed that,in the place navigation experiment,with the increase of training time,the escape latency of each group of mice decreased.Compared with the blank group,the escape latency and swimming distance of mice in the model group were obvious increased(P<0.05).Compared with the model group,the escape latency and the swimming distance of mice in the TCM group,the acupuncture group and the combination group were obvious decreased(P<0.05),and the changes in the combination of acupuncture and traditional Chinese medicine group were more obvious.In the spatial probe experiment,compared with the blank group,the frequency of crossing the platform in the model group were obvious decreased(P<0.05).Compared with the model group,the frequency of crossing the platform in the TCM group,the acupuncture group and the combination group were obvious increased(P<0.05),and the change in the combination of acupuncture and traditional Chinese medicine group was more obvious.5.After treatment,immunohistochemical staining technique test result showed that,compared with that in the blank group mice,the expression of Ch AT in the model group mice decreased significantly(P<0.05);compared with that in the model group mice,the expression of Ch AT in the TCM group,and the acupuncture group,as well as in the combination group increased significantly(P<0.05),and the increase in the expression by the combination group was comparatively more obvious.It was also observed that,compared with that in the blank group mice,the expression of Ach E in the model group mice increased significantly(P<0.05);compared with the model group mice,the expression of Ach E in the TCM group,and the acupuncture group,as well as in the combination group decreased significantly(P<0.05),and the decrease in the expression by the combination group was comparatively more obvious.6.After treatment,the Elisa test results showed that,compared with the blank group,the contents of A?,IL-1?,TNF-? in the model group were obvious increased(P<0.05).Compared with the model group,the contents of A?,IL-1?,TNF-? in the TCM group,the acupuncture group and the combination group were obvious decreased(P<0.05),especially in the combination group.Conclusion:1.The combination of Erchen Decoction and Taohong Siwu Decoction with acupuncture can improve the learning and memory ability of mice with mild cognitive impairment caused by arteriosclerosis.2.The combination of Erchen Decoction and Taohong Siwu Decoction with acupuncture can reduce the content of inflammatory cytokines in mice with mild cognitive impairment caused by arteriosclerosis,increase the expression of Ch AT,decrease the expression of Ach E.3.The combination of Erchen Decoction and Taohong Siwu Decoction with acupuncture treatment may affect the cholinergic system and inflammatory response in mice with mildcognitive impairment caused by arteriosclerosis.
