objective: The study of the regulatory network between the specific expression of Lnc RNA and m RNA gene function in acute and chronic wounds,provides a theoretical basis for exploring the mechanism of Huiyang and Yangyin.Methods: Gene chip technology was used to extract the total RNA of chronic refractory wounds and acute wounds,and then select the 2-fold difference and P <0.05 Lnc RNA and m RNA for differential expression analysis to obtain gene expression profiles.The KEGG pathway analysis of the m RNAs in the gene expression profile can find the most obvious pathways that are down-regulated and up-regulated,and then display the Pathway Map of the most obvious pathways respectively,so that significant m RNA indicators can be clearly obtained,and these significant m RNA indicators can be carried out q-PCR verification.After obtaining meaningful positive indicators,the Lnc RNA-m RNA co-expression network was obtained by intersecting with the differential Lnc RNA,and different Lnc RNAs were selected for functional verification based on the Lnc RNA-m RNA co-expression network.Results: Detection of gene expression profiles after differential analysis,502 up-regulated and 1204 down-regulated long-chain non-coding rnas and mrnas with different expressions were analyzed.MRNA analysis revealed621 up-regulated pathways and 505 down-regulated pathways,and KEGG pathway analysis revealed 10 up-regulated and down-regulated pathways.Through the Pathway Map display of the most obvious up-regulated and down-regulated pathways,8 significant m RNA indicators were selected:BMP2,IFNB1,IL6,IL18,PIK3 CB,SMAD7,SMAD9,and beta-actin,which were verified by q-pcr,and 5 significant positive indicators were obtained: BMP2,IL6,IL18,PIK3 CB and SMAD7 were intersected by the five positive indicators and differentially expressed lncrna-mrna to obtain the lncrna-mrna co-expression network,and different lncrnas were selected based on the lncrna-mrna co-expression network for functional verification.Conclusion: The 20 Lnc RNAs selected based on the Lnc RNA-m RNA co-expression network have different degrees of reports in various fields,providing directions and focus for the subsequent function and mechanism research,and exploring the mechanism of returning to Yang and nourishing yin.It provides a theoretical basis and provides new ideas for the study of traditional Chinese medicine to accelerate the healing of chronic refractory wounds.
